薏苡黑穗病拮抗菌的分离鉴定及其发酵条件优化

【摘要】:采用梯度稀释平板涂布法从土壤中分离并纯化细菌,通过平板对峙法、滤纸片法筛选出拮抗薏苡黑粉菌的细菌,经形态观察、16S rRNA基因序列分析鉴定菌株,并优化其发酵条件。结果表明,共分离得到拮抗菌株16个,其中拮抗菌菌株X-2对薏苡黑粉菌的抑制率相对最高,为66.7%,能够明显抑制病原菌的生长;经鉴定,菌株X-2为枯草芽孢杆菌(Bacillus subtilis),其最适发酵条件为温度34℃、pH值6.0、转速180 r/min、接种量4%、装液量200 m L/L。
[Abstract]:The bacteria were isolated and purified from soil by gradient dilution plate coating method. The antagonistic bacteria against Coix lachrymae were screened by plate confrontation method and filter paper method. The strains were identified by morphological observation and sequence analysis of 16s rRNA gene. The results showed that 16 antagonistic strains were isolated, among which X-2 had the highest inhibition rate to Coix lachryma-black powder bacteria, which was 66.7, which could inhibit the growth of pathogenic bacteria obviously.
