【摘要】:为了提高畜禽粪便腐熟的效率,试验采用平板扩散法和Folin-酚试剂比色法筛选耐热产蛋白酶芽孢杆菌菌株,并对其进行种属鉴定和产芽孢条件优化。结果表明:得到一株耐热产蛋白酶活性较高的芽孢杆菌菌株N62,初步鉴定为甲基营养型芽孢杆菌(Bacillus methylotrophicus);最适的培养基组成为玉米粉1.0%,黄豆饼粉1.0%,MgSO_4·7H_2O 0.02%,FeSO_4·5H_2O 0.02%;最适的培养条件为初始pH值7,装液量50 mL/250 mL,接种量8 mL,发酵时间72 h;优化后芽孢产率为95.0%。
[Abstract]:In order to improve the efficiency of fecal decay of livestock and poultry, the strain of Bacillus thermophilus was screened by plate diffusion method and Folin-phenol reagent colorimetry, and its species identification and spore production conditions were optimized. The results showed that a strain of Bacillus methylotrophicusum with high protease activity was obtained and identified as Bacillus methylotrophicus.The optimum culture medium was corn powder 1.0, soybean cake 1.0 7H_2O 0.02 and FeSO4 5H_2O 0.02; the optimum culture conditions were obtained. The initial pH value was 7, the volume of solution was 50 mL/250 mL, the inoculation amount was 8 mL, the fermentation time was 72 h, and the yield of spore was 95.0%.
