响应面分析法优化多功能菌B1514的产孢工艺

【摘要】:为研究多功能菌B1514的最佳产孢工艺,为其扩培、制剂的储存及使用提供参考,首先测定多功能菌B1514的生长曲线以初步确定其培养时间,然后采用单因素试验对影响多功能菌B1514产孢的接种量、初始p H值、装液量、转速、发酵温度和接种种龄6个因素进行筛选,筛选出具有显著性影响的主效应因素,再通过中心组合设计和响应面分析试验,以芽孢数量为响应值确定各显著影响因子的最佳水平,最后利用自动玻璃发酵罐对优化所得最佳培养条件进行验证。结果表明,最佳产孢工艺为接种量6%,初始p H值7.6,三角瓶装液量80 m L/500 m L,转速235 r/min,发酵温度36.5℃,接种种龄24 h,培养时间36 h,经验证芽孢数量达8.19×109CFU/m L,较优化前增加49%。
[Abstract]:In order to study the optimum sporulation technology of multifunctional bacteria B1514, and to provide reference for its expansion, storage and use, the growth curve of multifunctional bacteria B1514 was determined to determine its culture time. Then the single factor test was used to screen the inoculation amount, initial pH value, liquid loading amount, rotational speed, fermentation temperature and inoculation age of the multifunctional bacteria B1514, and the main effect factors were screened out. Through the central combination design and the response surface analysis test, the optimum level of the significant influence factors was determined with the number of spores as the response value. Finally, the optimum culture conditions were verified by using the automatic glass fermenter. The results showed that the optimum sporulation technology was inoculation volume of 6, initial pH value of 7.6, liquid volume of 80 mL / 500 mL, rotation speed of 235 r / min, fermentation temperature of 36.5 鈩,
