怀地黄内生细菌的分离鉴定及抗菌活性

【摘要】:从药用植物怀地黄的根中分离出32株内生细菌,并选择大肠杆菌、金黄色葡萄球菌、黑曲霉、地黄斑枯病和地黄根腐病5种菌作为供试菌株,采用点菌法、平板对峙法和菌饼法测定抗菌活性。结果显示,怀地黄中分离的32株内生菌对5种供试菌具有不同的抑制能力,其中有8株细菌对供试的大肠杆菌、金黄色葡萄球菌和黑曲霉都有抑制作用,显示出广谱的抗菌性;有2株细菌DH-10和DH-11对选取的5种供试菌都有抑制作用,且抗菌活性较强,具有生防菌开发潜力。所分离的活性内生菌通过16S rRNA基因序列分析,主要分布在芽孢杆菌属、假单胞菌属和大肠埃希氏菌属,其中对地黄斑枯病和根腐病都具有拮抗作用的内生菌主要集中在芽孢杆菌属中。
[Abstract]:32 strains of endophytic bacteria were isolated from the root of Radix Rehmanniae, and 5 strains of Escherichia coli, Staphylococcus aureus, Aspergillus Niger, Rehmannia blight and Rehmannia root rot were selected as the tested strains. The antimicrobial activity was determined by plate confrontation method and bacterial cake method. The results showed that 32 strains of endophytic bacteria isolated from Rehmannia glutinosa had different inhibition ability to 5 kinds of bacteria. Among them, 8 strains of bacteria had inhibitory effect on Escherichia coli, Staphylococcus aureus and Aspergillus Niger, which showed broad-spectrum antimicrobial activity. Two strains of bacteria DH-10 and DH-11 all had inhibitory effects on the five tested bacteria, and had strong antibacterial activity, which had the potential to develop biocontrol bacteria. The isolated active endophytic bacteria were mainly distributed in Bacillus, Pseudomonas and Escherichia coli by 16s rRNA gene sequence analysis. The endophytic bacteria which have antagonistic effect on both macular blight and root rot are mainly concentrated in the genus Bacillus.
