影响土壤中可溶性总氮、铵态氮和硝态氮测定条件的研究

【摘要】:为了筛选出测定土壤可溶性总氮、铵态氮和硝态氮的最优条件,土壤采集于内蒙古呼和浩特市西把栅乡的菜地,上一茬种植的是油麦菜;西把栅乡的农地,上一茬种植的是玉米;林地,采集于赛罕区树木园;皆为新鲜土壤作为实验材料。探索了用碱性过硫酸钾消解紫外分光光度法测定土壤溶解性总氮含量、用靛酚蓝比色法测定土壤铵态氮含量、用紫外分光光度法测定土壤硝态氮含量过程中,浸提温度和时间、震荡强度、过硫酸钾纯度、高压灭菌器消解的温度和时间、放气过程、降压过程、显色时间和温度等条件对测试结果的影响。通过方差分析结果表明和最终试验结果的含量对比,土壤可溶性总氮的最佳测试条件为:土壤浸提震荡温度22℃,转速160转/秒,浸提时间为60min,高压灭菌器消解温度122℃,时间40-50min,升压过程中不放气,人工放气后取出,药品选用进口过硫酸钾或国产过硫酸钾二次结晶体。测定土壤铵态氮含量最佳测试条件:土壤浸提温度为26℃-28℃,浸提时间为60min,震荡强度为160转/秒,显色时间为40min,显色温度为15℃。测定土壤硝态氮含量最佳测试条件:土壤浸提温度18℃-20℃,浸提时间为40min,震荡强度为250转/秒。
[Abstract]:In order to select the optimum conditions for the determination of soluble total nitrogen, ammonium nitrogen and nitrate nitrogen, the soil was collected in the vegetable field of Xijia Township, Hohhot City, Inner Mongolia. The farmland in Xichang Township was planted with corn in the last crop; the woodland was collected in the arboretum of Saihan region; all of them were fresh soil as experimental materials. The content of dissolved total nitrogen in soil was determined by alkaline potassium persulfate digestion ultraviolet spectrophotometry, the content of ammonium nitrogen in soil was determined by indigo blue colorimetry, and the extraction temperature and time were determined by ultraviolet spectrophotometry. The effects of shock intensity, purity of potassium persulfate, temperature and time of high pressure sterilizer digestion, degassing process, depressurization process, color reaction time and temperature on the test results. The results of variance analysis showed that the optimum test conditions of soil soluble total nitrogen were as follows: shaking temperature 22 鈩,
