黑曲霉YF1菌株对烟嘧磺隆的降解条件研究

【摘要】:黑曲霉(Aspergillus niger)YF1菌株是本实验室筛选得到的对烟嘧磺隆具有较强降解能力的菌株。为优化YF1菌株的降解条件,本试验分别对葡萄糖浓度、培养温度、初始pH和转速进行单因素试验,结果表明,YF1菌株降解烟嘧磺隆的最适条件为:葡萄糖浓度5g/L、培养温度30℃、初始pH值6.50和转速50r/min。本试验在研究其降解条件的同时,发现溶液的pH均出现下降,随后采用高效液相色谱检测草酸、L-苹果酸、α-酮戊二酸、乙酸、柠檬酸、D-苹果酸、琥珀酸的标准品,通过保留时间确定样品中的有机酸为草酸,但未检测出其它6种有机酸,因此,可以确定YF1菌株可以代谢葡萄糖产生草酸。本试验初步明确了黑曲霉YF1菌株对烟嘧磺隆的降解特性,为该菌株的应用奠定了基础。
