不同周期脉冲染料激光对兔耳创面上皮间充质交替对话及瘢痕形成的影响

【摘要】:目的:探讨脉冲染料激光(pulsed dye laser,PDL)不同治疗周期对兔耳创面上皮细胞向间质细胞转化的影响及该现象对瘢痕增生的作用。方法:新西兰大耳白兔12只建立兔耳腹侧增生性瘢痕模型,应用595nm Vbeam脉冲染料激光分别采用一周、二周、三周的治疗周期进行瘢痕的激光干预,在治疗前及治疗2、4、6、8周测量瘢痕厚度,治疗结束后取材行组织形态学观察,免疫组织化学方法测定各组上皮间充质转化(Epithelial-mesenchymal Transformation,EMT)正相关蛋白及负相关蛋白的表达情况。结果:兔耳各组模型均有瘢痕增生,EMT表面标记物表达阳性;PDL治疗结束后,一周治疗组及二周治疗组均表现出TGF-β1、Vim表达下调,Ecad表达上调,产生明显的瘢痕抑制效应,与对照组相比有显著差异;三周治疗组与对照组、一周治疗组和二周治疗组相比无明显差异。结论:PDL可能通过干预EMT的表达产生瘢痕抑制作用;治疗周期为两周较为适宜。
[Abstract]:Objective: to investigate the effect of pulsed dye laser (PDL) on the transformation from epithelial cells to interstitial cells in rabbit ear wound and the effect of this phenomenon on scar proliferation. Methods: the hypertrophic scar model was established in 12 New Zealand white rabbits. Laser intervention was performed with 595nm Vbeam pulsed dye laser for one week, two weeks and three weeks respectively. The thickness of scar was measured before treatment and 8 weeks after treatment. Histomorphology was performed after treatment. The expression of Epithelial-mesenchymal Transformation (EMTT) positive and negative correlation protein was detected by immunohistochemical method. Results: after the end of PDL treatment, the expression of EMT surface markers in each rabbit ear model showed a down-regulation of TGF- 尾 _ 1 Vim expression, which resulted in a marked scar inhibition effect in both the treatment group and the two-week treatment group, and the expression of TGF- 尾 _ 1 and Vim was down-regulated. There was no significant difference between the three-week treatment group and the control group, the one-week treatment group and the two-week treatment group. Conclusion the effect of chicatricial inhibition may be produced by intervention of the expression of EMT, and the treatment cycle is more suitable for two weeks.
