雌激素调控去势小鼠疼痛的炎症机制

【摘要】:目的·通过建立去势小鼠模型并给予外源性雌激素补充治疗,探索雌激素调控去势小鼠疼痛的炎症机制。方法·24只12周龄C57BL/6J雌性小鼠随机分为3组:假手术组(S组),手术+安慰剂对照组(P组),手术+雌激素替代治疗组(E组)。术后1周给予雌激素替代治疗,为期4周。分别在术前1日、术后3日及术后1、2、3、4、5周检测小鼠基础痛阈,包括机械痛阈缩足阈值(PWT)和热板痛阈缩足潜伏期(PWL);术后3日及5周检测小鼠血清炎症因子水平,包括IL-1β、IL-6、TNF-α;术后5周取小鼠脊髓腰膨大,Western blotting分析p38表达含量。结果·去势小鼠术后基础痛阈均较S组低,PWT、PWL分别在术后第1周、术后第3日起与S组差异有统计学意义(P0.01)。术后第3日,E组、P组炎症因子水平均较S组高,差异具有统计学意义(P0.01)。术后第5周,P组p38表达高于S组,差异具有统计学意义(P0.01)。给予雌激素补充治疗后,术后第4、5周,E组PWT、PWL均略有提高,与P组差异具有统计学意义(P0.01)。术后第5周E组炎症因子表达降低,与P组差异具有统计学意义(P0.01),但也仍然显著高于S组(P0.01)。术后第5周E组p38表达含量降低但仍高于S组,与P组差异具有统计学意义(P0.01),而与S组比较差异已无统计学意义(P0.05)。结论·雌激素可能通过p38 MAPK通路降低炎症因子水平,发挥对去势小鼠疼痛的调控作用。
[Abstract]:Objective to explore the inflammatory mechanism of estrogen regulating pain in ovariectomized mice by establishing ovariectomized mice model and giving exogenous estrogen supplementation therapy. Methods Twenty-four 12-week-old female C57BL/6J mice were randomly divided into three groups: sham operation group (group S), surgical placebo control group (group P) and estrogen replacement therapy group (group E). Estrogen replacement therapy was given 1 week after operation for 4 weeks. The basic pain threshold (PWTs) and the latent period of hot plate pain threshold (PWTs) were measured at 1 day before operation, 3 days after operation and 5 weeks after operation, and the levels of serum inflammatory factors in mice were measured at 3 and 5 weeks after operation. The expression of p38 was detected by Western blotting at 5 weeks after operation. Results the basic pain threshold of castrated mice was significantly lower than that of group S at 1 week and 3 days after operation respectively. There was significant difference between group S and group S (P 0.01). The level of inflammatory factors in group E was higher than that in group S on the 3rd day after operation, and the difference was statistically significant (P 0.01). The expression of p38 in P group was higher than that in S group at 5 weeks after operation, and the difference was statistically significant (P 0.01). After estrogen supplementation therapy, PWTG PWL in group E increased slightly at the 4th week after operation, which was significantly higher than that in group P (P 0.01). At the 5th week after operation, the expression of inflammatory factors in group E was significantly lower than that in group P (P 0.01), but still significantly higher than that in group S (P 0.01). At the 5th week after operation, the expression of p38 in group E was decreased but still higher than that in group S, and there was a significant difference between group E and group P (P 0.01), but there was no significant difference between group E and group S (P 0.05). Conclusion estrogen may reduce the level of inflammatory cytokines through p38 MAPK pathway and play a role in regulating pain in ovariectomized mice.
