核桃楸外果皮活性成分的提取分离与应用

核桃楸是我国十分重要的药源植物,其树皮、树叶及果实、外果皮均可入药,有显著的抗氧化、消炎、抗癌以及抑菌等作用。本文研究了核桃楸外果皮活性成分的提取分离、三萜类物质提取工艺的优化以及其生物活性的研究应用,为开发核桃楸更多的综合利用价值提供理论依据。采用体积分数80%的乙醇溶液对核桃楸外果皮浸提,并依次使用石油醚、氯仿和乙酸乙酯进行萃取。各萃取相经过薄层色谱和柱层析色谱分离后,利用气相色谱(GC)和气相色谱-质谱(GC-MS)检测各相中活性成分。石油醚萃取相洗脱剂为石油醚:乙酸乙酯=20:80分离出6种活性成分,氯仿萃取相洗脱剂为氯仿:乙酸乙酯=40:60分离出7种活性成分,氯仿萃取相洗脱剂为氯仿:乙酸乙酯=20:80分离出6种活性成分,乙酸乙酯萃取相洗脱剂为氯仿:甲醇=80:20分离出4种活性成分。在单因素实验的基础上,根据Box-Behnken中心组合法设计多因素试验,利用响应面法优化提取工艺条件,探究出乙醇体积分数、超声温度、超声时间和液料比对核桃楸外果皮总三萜提取率的影响,确定最佳提取工艺条件,以同样方法提取核桃楸叶总三萜作为对比。核桃楸外果皮中总三萜最佳提取条件为:乙醇体积分数8...
