槐米黄酮物质的提取及其抗氧化性研究

槐米是豆科植物国槐的干燥花蕾,广泛栽培于中国的各个省市、日本和韩国。槐米是一种珍贵的中草药,具有清热、凉血和止血作用。本试验以产自山西省夏县的槐米为试验原料,研究了槐米黄酮类化合物的提取工艺,槐米中多种黄酮物质的测定方法及其抗氧化性能。本文研究结果如下:本研究建立了定性和定量分析槐米提取物中六种黄酮化合物的高效液相色谱方法。实验条件为:流动相由0.1%磷酸水溶液(A)和乙腈(B)组成。使用前用45μm微孔滤膜过滤,并超声脱气。色谱柱Diamonsil TMC18柱(250mm×4.6mm,5μm),柱温为30℃,进样量为5μL,流速为1.0 mL/min。目标组分的检测波长为360 nm。流动相梯度洗脱条件如下:0-30 min,95-65%A;30-35min,65-95%A;35-50 min,95%A。结果表明该方法准确、有效,可用于槐米黄酮物质的质量控制。采用超声辅助提取槐米中的芦丁、烟草苷、水仙苷、山奈酚、异鼠李素、槲皮素和总黄酮。通过单因素试验和响应面法确定了最佳提取条件为:乙醇浓度为70%,时间30 min,温度61℃,液固比15.30mL/g。芦丁、烟草苷、水仙苷、山奈...
