生物检材中的毒品及其代谢物的气相色谱—质谱联用检测

【摘要】: 本课题建立了固相微萃取、固相萃取以及传统的液-液萃取结合气相色谱-质谱联用法分析了多种新型滥用药物(2C-B、尼美西泮、硝基安定、咪哒唑仑、曲马多、美沙酮和苯丙胺类)在生物体内的原体药物及其代谢产物的定性、定量检测方法。实验以Wistar大鼠和人为生物模型,口服上述药物,收集24小时内的尿液和提取生物组织(肝、脑、血液、毛发等)作为生物检材,利用固相微萃取法从生物检材中提取了曲马多、美沙酮,考察了固相微萃取是的萃取温度、萃取时间、pH以及离子强度等实验条件;固相萃取法提取了多种生物检材中的2C-B、尼美西泮、硝基安定、咪哒唑仑及其它们代谢产物;利用液-液提取法直接快速提取了检材中的曲马多和苯丙胺类滥用药物及其它们的代谢产物。结合气相色谱-质谱联用法定性或定量分析了上述多种滥用药物及其代谢产物,实验定量分析了尿液中的曲马多、美沙酮和毛发中苯丙胺类滥用药物,定性检测了2C-B、尼美西泮、硝基安定、咪哒唑仑、曲马多、美沙酮在生物体内的代谢产物,根据实验结果推断了相关滥用药物在生物体内的代谢途径,建立了检测方法和相关的数据库。建立的实验方法已经成功运用于实际工作中,并取得了良好的效果。
[Abstract]:This paper established a solid phase microextraction, solid phase extraction and conventional liquid-liquid extraction combined with gas chromatography-mass spectrometry analysis of a variety of new drug abuse (2C-B, nimetazepam and nitrazepam, midazolam, tramadol, methadone and amphetamines) qualitative parent drug and its metabolites in vivo the quantitative detection method. Experiments in Wistar rat and human biological model, the oral drug, urine was collected within 24 hours and the extraction of biological tissues (liver, brain, blood, hair, etc.) as biological samples by SPME extracted from biological samples of tramadol, methadone, effects of extraction temperature, solid phase micro extraction extraction time, pH and ionic strength conditions; solid-phase extraction of a variety of biological samples in 2C-B, nimetazepam and nitrazepam, midazolam and their metabolites; using liquid-liquid extraction The method of direct rapid extraction samples in the tramadol and abuse of amphetamine type drugs and their metabolites. Combined with GC-MS qualitative or quantitative analysis of the various abuse of drugs and their metabolites, the quantitative analysis of urine in tramadol abuse of amphetamine type drugs, methadone and hair in the qualitative detection of 2C-B, nimetazepam and nitrazepam, midazolam, tramadol, metabolites of methadone in organism, according to the experimental results that the metabolic pathways related to drug abuse in the organism, established the detection method and the related database. The established method has been successfully applied to the practical work, and have achieved good results.
