电击伤后心脏组织TIMP-1及a-Actin表达的实验研究

【摘要】: 目的目前电击伤(死)案件随着电力事业蓬勃发展呈逐年增加的趋势,法医学检案中用低压电来自杀或谋杀的案件也不少见。目前国内外法医学家在生前电击伤(死)和死后电击伤的鉴别及电损伤时间的推断等方面研究取得了一定进展,但仍没有取得重大突破。为了准确鉴别生前电击和死后电击伤以及损伤时间的准确推断,我们设计了该实验。 方法1.利用220V交流电(50Hz, 150-200 mA)电击Wister大白鼠胸部(心脏体表投影位置)30秒钟复制动物模型。2.电击后在不同时间(0分钟、30分钟、1小时、2小时、3小时、4小时、5小时、6小时、8小时、12小时、1天、3天、6天、12天)断颈处死;正常Wister大白鼠断颈处死后不同时间(0分钟、30分钟、2小时)电击(电击方法及时间等同前)。手术打开胸腔,取出心脏,PBS液冲洗后,将标本置于10%甲醛溶液中固定,作病理切片(5μm),通过HE染色观察其组织病理学改变;以TIMP-1、a-Actin为一抗标记不同时间的心脏组织,观察其分布情况。 结果:电击后0分钟至6小时,可见心肌细胞呈灶性的变性、坏死,部分心肌细胞呈现波纹状改变;部分心肌细胞胞膜横纹不清;胞浆凝聚、
[Abstract]:Objective at present, the electric injury (death) cases are increasing year by year with the rapid development of electric power industry, and the cases of suicide or murder with low-voltage electricity in forensic medical examination cases are not uncommon.At present, forensic scientists at home and abroad have made some progress in the identification of antemortem electric injury (death) and postmortem electric injury, and in the estimation of electric injury time, but no significant breakthrough has been made.In order to accurately identify the antemortem electric shock and postmortem electric shock injury as well as the exact estimation of injury time, we designed the experiment.Method 1.Wister rats were subjected to 220V AC 50Hz, 150-200mAshock to the chest (the projection position of the heart surface was used to reproduce the animal model for 30 seconds.After electric shock, 30 minutes (1 hour, 2 hour, 3 hour, 4 hour, 6 hour, 8 hour, 12 hour, 1 day, 3 days, 6 days and 12 days) were amputated and executed.Normal Wister rats were given electric shock at different time (30 minutes and 2 hours after neck amputation) (the method and time of electric shock were the same as before.After operation, the thoracic cavity was opened, the PBS solution was taken out, and the specimen was fixed in 10% formaldehyde solution. The histopathological changes were observed by HE staining, and the tissue of heart was labeled with TIMP-1a -Actin for different time, and the tissue was fixed in 10% formaldehyde solution, and the pathological changes were observed by HE staining.Observe its distribution.Results: from 0 minutes to 6 hours after electric shock, myocardial cells showed focal degeneration, necrosis, corrugated changes in some cardiomyocytes, unclear membrane lines in some cardiomyocytes, and cytosolic condensation.
