三重脑震荡鼠脑色氨酸羟化酶及酪氨酸羟化酶变化研究

【摘要】: 目的:建立三重脑震荡(multiple cerebral concussion,MCC)大鼠模型,观察MCC大鼠脑干色氨酸羟化酶(Tryptophan hydroxylase,TPH)与基底节和中隔区酪氨酸羟化酶(tyrosine hydroxylase,TH)表达变化情况,探讨与MCC大鼠认知行为障碍相关的色氨酸羟化酶和酪氨酸羟化酶变化的关系,以了解三重脑震荡鼠认知行为障碍的神经生物学基础。方法:采用自制金属单摆式闭合性脑损伤机械打击装置成功复制脑震荡大鼠模型48只;大鼠随机分为1天、2天、4天、8天、16天和24天6个三重脑震荡组(n=8)和1个正常对照组(Control,CON,n=8)。应用免疫印迹技术(Western Blotting)分别检测大鼠脑震荡损伤后第1、2、4、8、16、24天脑干色氨酸羟化酶(Tryptophanhydroxylase,TPH)以及基底节和中隔区酪氨酸羟化酶(Tyrosine hydroxylase,TH)的表达变化规律。结果:免疫印迹结果显示,脑干TPH表达自伤后1天起,呈逐渐上调趋势,到伤后第2天该酶的表达到最高峰,伤后2-8天的表达升高,与对照组相比较有显著性差异(p<0.05)。伤后16天,TPH表达逐渐下降,到伤后第24天基本已经回落到对照组的水平。基底节TH在伤后表达总体呈上升趋势,到伤后第4天该酶的表达到最高峰,与对照组相比较,伤后2-16天均有显著性差异(p<0.05)。中隔区TH在伤后表达总体呈上升趋势,伤后第1天开始上升,到伤后第4天该酶的表达到最高峰,8天和16天略有下降,24天再次升高,伤后第4天和第24天的表达与对照组相比较有显著性差异(p<0.05)。结论:1、三重脑震荡大鼠伤后脑干色氨酸羟化酶的表达升高。2、三重脑震荡大鼠伤后基底节、中隔区酪氨酸羟化酶的表达升高。3、根据本课题组之前的研究以及本次研究结果推测三重脑震荡后大鼠的认知缺陷可能与脑干TPH及基底节和中隔区TH的表达升高有关。
[Abstract]:Objective: to establish a rat model of multiple cerebral concussionMCCs with triple cerebral concussion, and to observe the expression of Tryptophan hydroxylase (Tryptophan hydroxylase) and tyrosine hydroxylase tyrosine hydroxylase (THH) in the basal ganglia and septum of MCC rats. To explore the relationship between the changes of tryptophan hydroxylase and tyrosine hydroxylase associated with cognitive behavioral disorders in MCC rats and to understand the neurobiological basis of cognitive behavioral disorders in rats with triple concussion. Methods: Forty-eight rats with cerebral concussion were randomly divided into 6 concussion group (n = 6) and a control group (n = 48) and a normal control group (n = 48), which were randomly divided into two groups: 1 day, 2 days, 4 days, 8 days, 16 days, and 24 days, 6 triple concussion groups. Western blotting technique was used to detect the expression of tryptophan hydroxylase (Tryptophan hydroxylase), tyrosine hydroxylase (Tyrosine hydroxylaseTHH) and tyrosine hydroxylase (Tyrosine hydroxylaseTH3) in brain stem of rats after cerebral concussion injury. Results: the results of Western blotting showed that the expression of TPH in the brain stem increased gradually from 1 day after injury to the peak on the second day after injury, and increased at 2-8 days after injury. There was a significant difference between the two groups in comparison with the control group (P < 0.05). The expression of TPH decreased gradually on the 16th day after injury, and decreased to the level of the control group on the 24th day after injury. The expression of th in basal ganglia showed a general upward trend after injury, and the expression of th reached its peak on the 4th day after injury. Compared with the control group, the expression of th in basal ganglia was significantly different from that of the control group on the 2-16 days after injury (P < 0.05). The expression of th in septal area showed a general upward trend after injury, which began to rise on the first day after injury, and increased again on the 4th day after injury to the peak at the peak of 8 days and 16 days after injury, and then increased again at 24 days after injury. On the 4th and 24th day after injury, there was significant difference between the control group and the control group (p < 0.05). Conclusion the expression of tryptophan hydroxylase in brain stem of rats with brain stem concussion was increased. The expression of tyrosine hydroxylase in septum was increased. According to our previous study and the results of this study, we speculated that the cognitive deficiency of rats after triple concussion might be related to the increased expression of TPH and th in basal ganglia and septum.
