P物质在过敏性休克家兔体内的表达及法医学意义

【摘要】: 目的:观察P物质在过敏性休克家兔胃、小肠、圆小囊组织中随时间变化的规律,探讨其在过敏性休克中的作用,试图为过敏性猝死的法医学鉴定提供可靠的形态学依据。 方法:30只家兔随机分为实验组(24只)和对照组(6只)。实验组采用多人混合血清致敏家兔,建立过敏性休克模型。休克不同时间段(猝死即刻、休克0.5h、休克1h、休克2h)提取家兔肺脏、胃、小肠、圆小囊。(1)冰冻切片,常规HE染色;(2)应用免疫组化(IHC)SP法进行P物质染色,BI-2000计算机图像分析系统进行图像分析,计算阳性积分光密度值(IOD);(3)应用原位杂交(ISH)对P物质进行染色,BI-2000计算机图像分析系统进行图像分析,计算IOD。 结果:(1)IHC结果显示:与对照组相比,实验组各小组胃、肠、圆小囊P物质蛋白表达明显增加,且随着休克时间的变化逐渐减弱。统计学方差分析结果为:胃组织F=198.008,肠组织F=56.796,圆小囊组织F=67.231,全部P值小于0.001,均有统计学意义。SNK检验结果为:胃组织各组之间两两比较均有差别;肠组织除对照组与休克2h组之间无统计学意义外,其余各组两两比较均有差别;圆小囊除对照组与休克2h组之间无统计学意义外,其余各组两两比较均有差别。(2)ISH结果显示:与对照组相比,实验组各小组胃、肠、圆小囊P物质mRNA表达明显增加,且随着休克时间的变化逐渐减弱。统计学方差分析结果为:胃组织F=67.506,肠组织F=72.117,圆小囊组织F=216.798,全部P值小于0.001,均有统计学意义。SNK检验结果为:胃组织除对照组与休克2h组之间无统计学意义外,其余各组之间两两比较均有差别;肠组织各组之间两两比较均有差别:圆小囊各组之间两两比较均有差别。 结论:过敏性休克时,家兔胃、小肠、圆小囊组织的P物质蛋白表达和mRNA表达明显增加,且随着休克时间的变化逐渐减弱。P物质可为过敏性休克的法医学鉴定提供客观的形态学依据和参考指标。
[Abstract]:Objective: to observe the changes of substance P in the stomach, small intestine and small capsule of anaphylactic shock rabbits, and to explore the role of substance P in anaphylactic shock in order to provide reliable morphological basis for forensic identification of sudden anaphylactic death. Methods 30 rabbits were randomly divided into experimental group (n = 24) and control group (n = 6). Anaphylactic shock model was established in the experimental group by sensitizing rabbits with mixed human serum. Frozen sections of rabbit lung, stomach, small intestine, circular vesicle were extracted from different periods of shock (instant of sudden death, 0.5 h of shock, 1 hour of shock, 2 hours of shock). Routine HE staining was used to carry out the image analysis of substance P staining with BI-2000 computer image analysis system. The positive integral optical density (IOD) was calculated by in situ hybridization (ISH). The computer image analysis system of substance P staining BI-2000 was used for image analysis and IOD was calculated. Results compared with the control group, the expression of substance P protein in stomach, intestine and capsule of the experimental group increased significantly, and gradually decreased with the change of shock time. The results of ANOVA were as follows: stomach tissue (FN) 198.008, intestinal tissue (FN) 56.796 (P < 0.001), small capsule tissue (FN) 67.231 (all P < 0.001). The results of SNK test were as follows: there were differences among gastric tissue groups. There was no significant difference in intestinal tissue between the control group and the 2h shock group, but there was no significant difference between the other groups except the control group and the 2h shock group. The results showed that compared with the control group, the expression of substance P mRNA in the stomach, intestine and small capsule of the experimental group was significantly increased, and gradually weakened with the change of shock time. The results of ANOVA were as follows: stomach tissue (67.506), intestinal tissue (FN) 72.117, round capsule (FN) 216.798 (P < 0.001). The results of SNK test showed that there was no significant difference between gastric tissue and control group and 2 h shock group. There were differences between the other groups, and between the intestinal tissue groups and the small sac groups. Conclusion: during anaphylactic shock, the expression of substance P protein and mRNA in the stomach, small intestine and small capsule of rabbits were significantly increased. With the change of shock time, substance P can provide objective morphological basis and reference index for forensic identification of anaphylactic shock.
