新的STR遗传标记及荧光标记复合扩增的法医学应用研究

【摘要】:目的 本课题旨在寻找应用于法医的STR基因座并建立D20S601、D6S474、D6S2418荧光标记复合扩增体系,按照美国DNA分析方法技术工作组(The Technology Working Group on DNA Analysis Methods,TWGDAM)的指导方案进行法医学实用性研究。方法 对5个新的STR基因座进行群体遗传学研究,并对D20S601、D6S474、D6S2418基因座原始引物进行了改进,利用传统复合扩增技术扩增,ABI PRISM~(?)310基因分析仪对产物进行检测;用混合样本、不同的组织、不同的介质、常见的动物在不同的模板量、不同的温度条件下对复合扩增系统进行测试,用实际案例进行验证。结果 获得5个STR基因座的基因频率、个人识别率、非父排除率等群体遗传学数据;通过采用传统的复合扩增的方法,建立了D20S601、D6S474、D6S2418荧光标记复合扩增
[Abstract]:Objective the purpose of this study was to search for the STR loci used in forensic medicine and to establish a multiplex amplification system for D20S601, D6S474, D6S2418 fluorescent labeling. According to the (The Technology Working Group on DNA Analysis Methods, of the DNA analysis technique working group of the United States, TWGDAM) is used to study the practicability of forensic medicine. Methods five new STR loci were studied in population genetics. The primordial primers of D20S601D6S474nD6S2418 locus were improved. The products were amplified by traditional multiplex amplification technique and detected by ABI PRISM~ (?) 310 gene analyzer. The compound amplification system was tested with mixed samples, different tissues, different media, common animals under different template quantity and different temperature, and verified by actual cases. Results the population genetic data of 5 STR loci were obtained, such as gene frequency, personal identification rate, non-paternal exclusion rate, etc. By using the traditional multiplex amplification method, the fluorescent multiplex amplification of D20S601D6S474hD6S2418 was established.
