法医DNA分析若干问题的研究

【摘要】:目的对6个短串联重复序列(Short Tandem Repeats,STR)基因座在成都汉族群体中的遗传多态性及其种属特异性进行研究,探讨其在法医学中的应用价值,并为法医DNA分型提供新的候选STR遗传标记。 方法 用Chelex法从110名成都汉族无血缘关系个体的新鲜血以及14种常见动物组织中提取DNA。从GDB数据库中查出6个四核苷酸重复序列D4S2366、D4S2367、D6S474、D6S1281、D2S1396和D20S601,按数据库提供的序列合成引物,经PCR扩增、非变性聚丙烯酰胺凝胶电泳和硝酸银染色技术检测,对110名成都汉族无血缘关系个体的6个STR基因座进行DNA分型,应用POWERSTATS软件对分型结果进行统计分析,获得各基因座基因频率及法医遗传多态性数据。应用所合成的引物对14种常见动物DNA进行PCR扩增和电泳检测,对6个STR基因座的种属特异性进行研究。 结果 D4S2366、D4S2367、D6S474、D6S1281、D2S1396和D20S601基因座在成都汉族群体中等位基因个数分别为7、7、6、7、5和7;观察杂合度分别为0.802、0.708、0.770、0.627、0.542和
[Abstract]:Objective to study the genetic polymorphism and species specificity of six short tandem repeat (Short Tandem Repeats,STR) loci in Chengdu Han population, and to explore their application value in forensic medicine. And to provide a new candidate STR genetic marker for forensic DNA typing. Methods Chelex method was used to extract DNA. from fresh blood and 14 common animal tissues from 110 unrelated individuals of Chengdu Han nationality. Six tetranucleotide repeats D4S2366, D4S2367, D6S474, D6S1281, D2S1396 and D20S601 were identified from GDB database. Primers were synthesized according to the sequences provided by the database, amplified by PCR, detected by non-denatured polyacrylamide gel electrophoresis and silver nitrate staining. Six STR loci of 110 unrelated individuals in Chengdu Han nationality were genotyped by DNA. The results were statistically analyzed by POWERSTATS software, and the genetic polymorphism data of each locus and forensic genetic polymorphism were obtained. The PCR amplification and electrophoretic detection of DNA of 14 common animals were carried out by using the synthesized primers, and the species-specificity of 6 STR loci were studied. Results the number of alleles in D4S2366-D4S2367-D6S474FN D6S1281D2S1396 and D20S601 locus in Chengdu Han population were 70.7080.7270.542 and 0.8020.7080.7080.7270.542, respectively.
