新Y染色体STR基因座遗传多态性研究

【摘要】: 目的通过对DYS713、DYS723 Y-STR基因座在山西汉族群体的研究,获得各基因座的基因频率分布情况,为群体遗传学、法医学个体识别及亲子鉴定提供计算数据,对其在法医学上的应用进行可行性探讨。 方法从GDB数据库中寻找DYS713、DYS723 Y-STR基因座;用PCR方法扩增,电泳分型;对其所有等位基因测序,构建等位基因分型标准物;按照ISFG原则命名各等位基因。 结果(1)在本研究的群体样本中,DYS713、DYS723基因座分别观察到6、5个等位基因,其基因变异度分别为0.7370、0.6618,其个人识别能力和非父排除率分别为0.7370、0.6618。(2)20例女性样本DNA无扩增产物。(3)同一尸体血液、器官组织检测结果分型一致。(4)20例两代家系观察未见突变。 结论DYS713、DYS723基因座在山西汉族男性人群中均有较好的多态性,可用于法医学的混合斑分析和父系亲属间的亲权关系鉴定,对法医学应用、人类遗传学等研究均具有重要价值。
[Abstract]:Objective to obtain the frequency distribution of DYS713,DYS723 Y-STR loci in Shanxi Han population, and to provide data for population genetics, forensic individual identification and paternity identification. The feasibility of its application in forensic science was discussed. Methods DYS713,DYS723 Y-STR loci were obtained from GDB database, amplified by PCR and typed by electrophoresis. All alleles were sequenced to construct the allelic typing standard, and the alleles were named according to ISFG principle. Results (1) in the population samples of this study, 6 and 5 alleles were observed at the DYS713,DYS723 locus, and their genetic variations were 0.7370,0.6618,respectively. The individual recognition ability and the non-paternal exclusion rate were 0.7370 and 0.6618 respectively. (2) 20 female samples had no DNA amplification products. (3) the results of organ and tissue tests were consistent in the blood of the same cadaver. (4) No mutation was observed in 20 pedigree of two generations. Conclusion DYS713,DYS723 locus has good polymorphism in male population of Han nationality in Shanxi province. It can be used for mixed spot analysis of forensic science and paternity identification between paternal relatives. It is of great value for forensic application and human genetics research.
