不同重金属修复技术对铅污染下瞿麦品质的影响

【摘要】:近年来,中药材瞿麦(Dianthus superbus L.)的抗肿瘤作用受到普遍关注,其需求量逐渐增大。但瞿麦的道地产区河北、内蒙古等地的土壤均有不同程度的铅含量超标问题,使铅在种植的药材中累积,进一步影响临床医治效果。目前,国内外常用化学、生物共生、联合共生等方法来进行土壤修复,但对于是否会影响种植植物的品质却鲜有报道。本文通过盆栽实验研究了接种丛枝菌根真菌(AM真菌)、种植共生植物(三叶草)、分别添加重金属螯合物EDTA和磷肥磷酸钙对铅污染下瞿麦生长、品质及药效的影响,为科学种植中药材提供实践依据。主要研究结果如下:1.除添加EDTA外,其他方法均能阻碍瞿麦对土壤中铅的吸收,使铅含量显著降低,在AM真菌、三叶草与瞿麦共生时,瞿麦中的铅含量达到最低,降至1.29 mg·g-1。与对照组相比,有AM真菌参与的修复技术对瞿麦生长的改善最为显著。2.运用响应面法优化了瞿麦中大黄素的提取条件,最终确定了瞿麦中大黄素提取的最优条件为:液固比61倍,加入1 mL质量分数为8%的盐酸,在40℃时超声30 min。并测得在与AM真菌共生时,瞿麦中活性成分累计最多,大黄素含量为6.5 mg·g~(-1)。3.使用MTT法测定瞿麦的石油醚提取物对子宫颈癌细胞系Hela细胞和人胃癌细胞株MGC-803的抑制作用。实验表明,瞿麦石油醚提取物对Hela细胞的抑制率更显著(EDTA组除外),且植物体内重金属的含量与肿瘤细胞抑制率也有一定的相关性。
[Abstract]:In recent years, Dianthus superbus L. The anti-tumor effect of 1% has received widespread attention, and its demand is increasing gradually. However, the lead content in the soils of Hebei, Inner Mongolia and other places in the true-producing areas of Qumai was higher than the standard, which caused the accumulation of lead in the cultivated medicinal materials and further affected the clinical therapeutic effect. At present, chemical, biological symbiosis, combined symbiosis and other methods are commonly used to repair soil at home and abroad, but there are few reports on whether it will affect the quality of planting plants. The effects of inoculation of arbuscular mycorrhizal fungi (Amycorrhizal mycorrhizal fungi) on the growth, quality and efficacy of diqu wheat under lead pollution were studied in this paper. The effects of chelate EDTA and calcium phosphate fertilizer on the growth, quality and efficacy of diqu wheat under lead pollution were studied in this paper. To provide practical basis for scientific cultivation of traditional Chinese medicine. The main results are as follows: 1. In addition to the addition of EDTA, all the other methods could inhibit the absorption of lead in soil by diqu wheat, and make the lead content decrease significantly. When AM fungi, clover and cu wheat symbiosis, the lead content in diqu wheat reached the lowest level, to 1.29 mg g -1. Compared with the control group, the improvement of diqu wheat growth with AM fungi was the most significant. 2. 2. The extraction conditions of emodin from diqu wheat were optimized by response surface method.
