西藏血蜱对环境变化的生理响应

【摘要】:西藏血蜱Haemaphysalis tibetensis Hoogstraal,1965仅分布于西藏和甘肃,是青藏高原地区重要的病原菌载体。目前已对西藏血蜱形态学、种群动态、生活史和分子系统学等方面做了研究,但有关其对环境的生理适应研究尚属空白。本研究通过测定环境变化后西藏血蜱雌蜱体内水分和抗寒物质含量的变化分析其对环境变化的生理响应;通过探讨西藏血蜱体内抗氧化酶活性和膜脂过氧化物含量的变化分析其对环境胁迫的适应对策,为评价西藏血蜱的特殊地理分布和对环境的耐受性提供科学的依据,主要结果如下:1.水分:与野外种群相比,实验种群体内自由水含量从55.65%上升至56.96%,虽有上升趋势,但无显著差异。2.抗寒物质:与野外种群相比,实验种群体内糖原、脂肪和可溶性蛋白含量均显著升高(P0.05)。糖原含量从8.62 mg/g上升至11.16 mg/g;脂肪含量从16.39%上升至21.66%;可溶性蛋白含量从1.06 gprot/l上升至1.16 gprot/l。实验种群血淋巴甘油含量显著降低(P0.05),从12.21μg/ml下降至10.81μg/ml。3.抗氧化酶和丙二醛(MDA):随西藏血蜱的发育,抗氧化酶活性和丙二醛含量均增加,雌蜱时期最高。与野外种群相比,实验种群体内保护酶活性和MDA含量均显著增加(P0.05)。超氧化物歧化酶(SOD)活性从20.27 U/mgprot增至26.06 U/mgprot;过氧化氢酶(CAT)活性从16.9 U/mgprot增至20.06 U/mgprot;过氧化物酶(POD)活性从6.96 U/mgprot增至9.40 U/mgprot;MDA含量从12.35 nmol/mgprot增至14.48nmol/mgprot。谷胱甘肽硫转移酶(GSTs)活性从87.67 U/mgprot增至90.66 U/mgprot,虽有上升趋势,但差异不显著。4.温度对抗氧化酶和丙二醛的影响:以22°C西藏血蜱雌蜱体内抗氧化酶活性和MDA含量作为对照,随着温度的降低SOD活性逐渐升高,除20℃外SOD活性与对照相比差异均显著(P0.05);在-5℃和0℃时CAT活性显著升高,在10℃时其活性显著降低(P0.05);在-5℃、0℃和5℃时POD活性显著升高(P0.05);在-5℃时GSTs显著升高,在5℃、10℃和15℃时其活性显著降低(P0.05);在5℃、10℃、15℃时MDA含量显著降低(P0.05)。其余温度条件下,抗氧化酶活性和MDA含量虽有变化,与对照相比差异不显著。
[Abstract]:Haemaphysalis tibetensis Hoogstraalus is only distributed in Tibet and Gansu, and is an important pathogen carrier in Qinghai-Tibet Plateau. The morphology, population dynamics, life history and molecular phylogeny of Haemaphysalis Xizang have been studied. However, the study on physiological adaptation to the environment was still blank. The physiological response to environmental changes was analyzed by measuring the changes of water and cold-resistant substances in female tick of Haemaphysalis Tibet after environmental change. The changes of antioxidant enzyme activity and membrane lipid peroxide content in Haemaphysalis Xizang were studied to analyze their adaptation to environmental stress, which provided a scientific basis for evaluating the special geographical distribution and tolerance to the environment of Haemaphysalis Tibetan Haemaphysalis. The main results were as follows: water: compared with the wild population, the free water content in the experimental population increased from 55.65% to 56.96, although there was an upward trend, there was no significant difference between them. 2. Cold resistant substances: compared with the field population, the glycogen in the experimental population, The content of glycogen increased from 8.62 mg/g to 11.16 mg / g; the content of fat increased from 16.39% to 21.66; the content of soluble protein increased from 1.06 gprot/l to 1.16 g / L; the content of glycerol in hemolymph decreased significantly from 8.62 mg/g to 11.16 mg / g. 12.21 渭 g / ml decreased to 10.81 渭 g / ml.
