竹叶椒果实精油的提取工艺及功能性研究

【摘要】:本文以芸香科花椒属植物竹叶椒的果实为材料对其精油提取工艺以及功能性进行了相应研究,主要结果如下:1.采取水蒸气蒸馏法,通过单因素及正交试验对竹叶椒果实精油进行提取工艺优化,得出最佳提取工艺为料液比1:30,蒸馏时间100 min,微波辅助时间2.0 min,在最佳提取工艺条件下竹叶椒果实精油得率为4.01%。其中微波辅助时间对竹叶椒果实精油得率影响最大,其次为蒸馏时间。2.采用GC-MS对竹叶椒果实精油进行成分分析,通过分析鉴定出25种物质,主要为稀类物质和醇类物质,其中主要成分有三种,分别为桉叶油醇(36.19%)、和萜品烯(21.31%)和3-蒈烯(8.45%)。3.选取了大肠杆菌、普通变形杆菌、产气肠杆菌3种革兰氏阴性细菌,枯草芽孢杆菌、胶质芽孢杆菌、蜡状芽孢杆菌、苏云金芽孢杆菌、金黄色葡萄球菌5种革兰氏阳性细菌,以及酵母菌、根霉、米曲霉、曲霉、尖孢镰刀菌5种真菌研究竹叶椒果实精油的抑菌效果,结果表明,竹叶椒果实精油对细菌的抑制具有广谱性,且对革兰氏阳性细菌的抑制效果强于革兰氏阴性细菌,对真菌的抑制作用不明显。4.用提取的竹叶椒果实精油为材料,采用两种熏香方式三种剂量处理小鼠,结果表明两种方式的三个剂量对小鼠超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)均有提升作用,对丙二醛(MDA)含量有降低效果,且随着剂量的增加都呈现良好的线性;中剂量和高剂量均达到显著性差异,说明在研究的剂量范围内,竹叶椒果实精油对小鼠有很好的抗氧化效果。抗疲劳试验中,竹叶椒果实精油两种处理方式对小负重游泳时间和爬杆时间均有增加作用,且中剂量和高剂量效果显著,说明对小鼠抗疲劳有很好的效果。通过观测小鼠进食后2小时内的睡眠率可以发现,精油熏香处理可以增加小鼠睡眠率,且随着时间和剂量的增加效果更明显。从处理方式来看,在试验剂量相同的情况下在抗氧化效果、抗疲劳程度、增进睡眠方面加湿熏蒸法都比干热熏蒸法效果好。5.通过上述提取工艺和处理方式的结果,总结出竹叶椒果实精油的微波辅助熏香装置,针对广大社会人群,具有结构简单,控制方便,条件温和的特点。
[Abstract]:In this paper, the extraction technology and function of essential oil from the fruit of bamboo leaf pepper of Rutaceae were studied. The main results are as follows: 1. The extraction process of essential oil from bamboo leaf pepper fruit was optimized by single factor and orthogonal test. The optimum extraction conditions were as follows: ratio of material to liquid 1: 30, distillation time 100 min, microwave assistant time 2.0 min. Under the optimum extraction conditions, the yield of essential oil of bamboo leaf pepper fruit was 4.01%, among which microwave assisted time had the greatest effect on the yield of essential oil of bamboo leaf pepper fruit. Secondly, distilling time. 2. The essential oil of bamboo leaf pepper fruit was analyzed by GC-MS, and 25 kinds of substances were identified, which were mainly dilute substances and alcohols, three of which were main components. E. coli, Proteus vulgaris, Enterobacter aerogenes, Bacillus subtilis, Bacillus glia, Bacillus cereus, Bacillus thuringiensis, Bacillus cereus, Bacillus thuringiensis, Bacillus cereus, Bacillus thuringiensis, Enterobacter aerogenes, Bacillus subtilis, Bacillus cereus, Bacillus thuringiensis, Bacillus cereus, Bacillus thuringiensis, Bacillus thuringiensis, Bacillus cereus, Bacillus thuringiensis. Five Gram-positive bacteria of Staphylococcus aureus, as well as yeast, Rhizopus, Aspergillus oryzae, Aspergillus sp., Fusarium oxysporum. The inhibitory effect of essential oil on bacteria was broad spectrum, and the inhibitory effect on Gram-positive bacteria was stronger than that on Gram-negative bacteria, and the inhibitory effect on fungi was not obvious .4.The extracted essential oil of bamboo leaf pepper fruit was used as material. Two kinds of incense fumigation and three doses of fumigation were used to treat mice. The results showed that the three dosages of two ways could enhance the activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and decrease the content of malondialdehyde (MDA) in mice. With the increase of dose, there was a good linearity and significant difference between middle dose and high dose, which indicated that the essential oil of bamboo leaf pepper fruit had a good antioxidation effect on mice in the range of the study, and the anti-fatigue test showed that in the anti-fatigue test, the essential oil of the fruit of Phyllostachys bungeanum had a good antioxidation effect. The two treatments of the essential oil of the fruit of bamboo leaf pepper increased the swimming time and the climbing time of small load, and the effect of middle and high dose was remarkable. By observing the sleep rate of mice within 2 hours after eating, it was found that essential oil fumigation could increase the sleep rate of mice. And with the increase of time and dose, the effect is more obvious. From the way of treatment, when the test dose is the same, the anti-oxidation effect, the degree of anti-fatigue, The humidification fumigation method is better than the dry heat fumigation method in improving sleep. 5. The microwave assisted fumigation device for the essential oil of bamboo leaf pepper fruit is summarized through the results of the above extraction technology and treatment method. Easy control and mild conditions.
