新型近红外荧光染料的合成、光谱测试与生物成像应用

【摘要】:荧光染料在生物医学领域有着广泛的应用,如检测、标记、光动力治疗、光热消融治疗等。近红外光的波长比普通可见光波长更长,其瑞利散射效应更小,具有更好的组织穿透性;此外近红外光的光毒性低,光热性好,受到生物组织背景信号干扰小,650-900 nm的波段被称为生物成像的"近红外窗口",因此近红外荧光染料的开发对生物医学的发展起着极大的推进作用。然而实际可用的近红外染料数量少,且有着诸多的缺点,例如菁染料化学稳定性和光稳定性较差,氟硼二吡咯染料溶解性难以满足要求。目前大多数近红外染料的最大发射波长集中在650-820 nm范围内,处于820-900 nm范围内的染料鲜有报道,此波长范围内的染料有着重要的学术研究和实际应用价值。本课题组近期在近红外染料的设计与合成中取得了突破,首次报道了二苯并碳罗丹明的母核结构;基于该母核的染料ECX其最大吸收波长可以达到880 nm,ECX有着比该波长范围内其他染料更为优越的化学和光学性质;ECX具有良好的生物兼容性且成功应用到细胞成像中。该课题正是在此工作基础上,对新开发的染料母核进行修饰,合成了一系列新型近红外荧光染料ECY;随后对它们的光物理性质和化学稳定性进行测试,最后把ECY染料连接到靶向基团上实现了对肿瘤细胞的识别,最终利用荧光成像和光声成像技术对肿瘤组织进行成像。本课题验证了 ECY染料在近红外区域的吸收和发射,同时证明了 ECY染料具有良好的化学和光学性质,具备生物兼容性,在荧光成像和光声成像中作为造影剂有可观的应用前景。
[Abstract]:Fluorescent dyes are widely used in biomedical fields, such as detection, labeling, photodynamic therapy, photothermal ablation and so on. The wavelength of near-infrared light is longer than that of ordinary visible light, its Rayleigh scattering effect is smaller, and it has better tissue penetration. The band of 650-900 nm which is interfered by the background signal of biological tissue is called the "near infrared window" of biological imaging, so the development of near infrared fluorescent dyes plays an important role in the development of biomedicine. However, the practical use of near infrared dyes is small and has many shortcomings, such as the poor chemical stability and photostability of cyanine dyes, the solubility of fluorinated boron dipyrrole dyes is difficult to meet the requirements. At present, the maximum emission wavelengths of most near infrared dyes are concentrated in the range of 650-820 nm, and the dyes in the range of 820-900 nm are rarely reported. The dyes in this wavelength range have important academic research and practical application value. Recently, our team has made a breakthrough in the design and synthesis of near infrared dyes, and reported for the first time the parent nuclear structure of dibenzo carbon rhodamine. The maximum absorption wavelength of ECX based on the parent nucleus can reach 880 nm. ECX has better chemical and optical properties than other dyes in this wavelength range. ECX has good biological compatibility and has been successfully applied to cell imaging. On the basis of this work, a series of novel near infrared fluorescent dyes ECY were synthesized by modifying the newly developed dye mother nuclei, and their photophysical properties and chemical stability were tested. Finally, the ECY dye was connected to the target group to realize the recognition of tumor cells, and finally the tumor tissue was imaged by fluorescence imaging and photoacoustic imaging. In this paper, the absorption and emission of ECY dyes in near infrared region are verified, and it is proved that ECY dyes have good chemical and optical properties and biological compatibility. It has a promising future as a contrast agent in fluorescence imaging and photoacoustic imaging.
