绞股蓝皂苷缓解油酸诱导的HepG2细胞脂肪堆积的作用

【摘要】:随着我国经济的快速发展,我国人民的生活水平得到了飞速的提升。与过去相比,我国人民饮食喜欢和饮食结构发生了巨大的变化,肉制品成为饭桌上的常客,并且随着生活节奏的加快,外食成了目前人们的主要就餐方式。这一系列的改变都导致人民饮食摄取的油脂大幅度增加,高血脂、心脑血管疾病的发生率也在大大的增加。血脂异常是导致这一系列疾病的重要因素,它是指人体内甘油三酯、总胆固醇、高密度脂蛋白和低密度脂蛋白等一系列指标异常。绞股蓝(Gynostemma pentaphyllum Thunb.Makion)具有改善脂质代谢、抗癌防衰、滋补保健、增强体质等多种功能,有着"南方人参"之美誉。绞股蓝皂苷是它的主要活性物质,具有降血糖、降血脂、保护心脑血管、抗肿瘤、抗氧化及抗炎等生理活性,越来越受到国内外学者的关注。本课题以降血脂有效成分绞股蓝达玛烷型四环三萜类化合物为研究对象,通过建立体外HepG2肝细胞脂肪堆积模型,评价绞股蓝皂苷清除肝细胞脂肪堆积的能力。主要的研究内容和结论如下:采用MTT法,筛选了 5个绞股蓝皂苷对HepG2细胞活力的影响,确定化合物SL-1和SL-2为合适的样品,并且明确合适的给药浓度范围为10μg/ml-100 μg/ml。通过反复柱层析法,从绞股蓝总皂苷中快速分离制备绞股蓝皂苷SL-1和SL-2。利用油酸诱导建立HepG2细胞脂肪堆积模型,从细胞内脂滴形态、总胆固醇(TC值)、甘油三酯(TG值)、高密度脂蛋白胆固醇(HDL-c值)、低密度脂蛋白胆固醇(LDL-c值)等方面评价绞股蓝皂苷的降脂作用。试验结果表明,绞股蓝皂苷SL-1和SL-2均可明显减轻细胞内脂质堆积现象,显著降低细胞中TG、TC含量,其中100μg/ml绞股蓝皂苷SL-1对TG、TC的清除率分别为达54.8%和52.7%;100 μg/ml绞股蓝皂苷SL-2对TG、TC的清除率分别为达78.3%和80.2%;并且高浓度的SL-1和SL-2对HDL和LDL也有很好的影响作用。由此可见,两种绞股蓝皂苷均具有良好的体外降脂活性,对脂肪肝有较好的预防效果。
[Abstract]:With the rapid development of our economy, our people's living standards have been rapidly improved. Compared with the past, the Chinese people's diet and diet structure have undergone great changes, meat products have become frequent guests on the table, and with the acceleration of life rhythm, external food has become the main way for people to eat at present. This series of changes have led to a significant increase in people's dietary fat intake, hyperlipidemia, cardiovascular and cerebrovascular diseases. Dyslipidemia is an important factor leading to this series of diseases. It refers to a series of abnormal indexes such as triglyceride, total cholesterol, high density lipoprotein and low density lipoprotein. Gynostemma pentaphyllum (Gynostemma pentaphyllum Thunb.Makion) has many functions, such as improving lipid metabolism, anticancer and anti-aging, nourishing health care, strengthening physique and so on. Gynostemma pentaphyllum saponins are the main active substances, which have the physiological activities of lowering blood sugar, lowering blood lipids, protecting heart and cerebrovascular vessels, anti-tumor, anti-oxidation and anti-inflammation and so on, which are paid more and more attention by scholars at home and abroad. The aim of this study was to evaluate the ability of gynostemma pentaphyllum saponins (Gypenosides) to scavenge lipid accumulation in hepatocytes by establishing an in vitro HepG2 hepatocyte fat accumulation model. The main contents and conclusions were as follows: by MTT method, the effects of five gynostemosides on the viability of HepG2 cells were screened, and the suitable samples of SL-1 and SL-2 were determined, and the suitable concentration range was 10 渭 g/ml-100 渭 g / ml. Rapid separation and preparation of SL-1 and SL-2. from total saponins of gynostemma pentaphyllum by repeated column chromatography The lipid accumulation model of HepG2 cells was established by oleic acid induction. Total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-c) and low density lipoprotein cholesterol (LDL-c) were used to evaluate the antilipidemic effect of gynostemma pentaphyllum saponins. The results showed that both ginsenoside SL-1 and SL-2 could significantly reduce the accumulation of lipid and the content of TG,TC in the cells. The clearance rates of 100 渭 g/ml gynosaponin SL-1 to TG,TC were 54.8% and 52.7 渭 g/ml, respectively, and the clearance rates of SL-2 to TG,TC were 78.3% and 80.2%, respectively, and the high concentrations of SL-1 and SL-2 also had a good effect on HDL and LDL. It can be seen that both gynostemma pentaphyllum saponins have good lipid-lowering activity in vitro and have a better preventive effect on fatty liver.
