丙泊酚脂肪乳注射液处方工艺优化及质量评价

【摘要】:丙泊酚(propofol)是一种静脉注射用麻醉药,由于其作用快、易控制等特点,其在临床的应用非常广泛,本课题选用丙泊酚为主药通过现代化的制备工艺,对丙泊酚脂肪乳注射液进行研究。建立了丙泊酚和有关物质的高效液相色谱检测方法,该方法灵敏度高、专属性好,适用于丙泊酚及有关物质的分析检测,采用十八烷基硅烷键合硅胶作为填充剂,柱温控制在30℃;分别采用275、254nm作为紫外检测波长。流动相A为磷酸二氢钠0.2%的水溶液,pH值为3.0,流动相B:乙腈,采用梯度程序洗脱法,流速为1.0 ml/min。在上述拟定的色谱条件下,有关物质A和B均能与主峰丙泊酚得到良好分离,且相邻峰之间的分离度超过1.5。同时采用蒸发光作为检测器,建立了溶血磷脂酰胆碱的高效液相色谱检测方法,该方法也具有灵敏度、专属性好的特点。通过单因素试验选择了 1.2%磷脂和0.05%油酸用量,选择了大豆油和中链甘油三酯比例为1:1的混合油作为油相载体。通过正交实验设计来优化制备工艺,最终选择高剪切乳化温度控制在70℃;NaOH选择在初乳制备前加入,高压均质压力为300bar,均质温度控制在60℃等工艺条件制备了丙泊酚脂肪乳注射液,并通过电子射镜和粒径分布等进行表征。在实验室处方和制备工艺基础上进一步进行中试工艺的优化,使用1OObar压力均质替代实验室的高剪切和均质混合来进行初乳的制备,选择了过滤压力≤40Kpa,以减少大于1μm的不溶性微粒数量;灌装充氮压力为30Kpa时,其灌装后的氧气残余量为3.0±0.4%;灭菌条件为121℃,12min,其中当工业蒸汽压力≥0.3Mpa,冷却水压力=0.2Mpa时,在保证F0值≥12的条件下既能缩短升温和降温时间又能得到较低的破损率。采用优化的中试生产工艺制得了丙泊酚脂肪乳注射液,其平均粒径为245.2±10nm,其它各项质量指标均符合要求。对丙泊酚脂肪乳注射液中试工艺生产的样品进行了影响因素试验、加速试验和长期试验,测定了 pH值、渗透压摩尔浓度、丙泊酚含量、有关物质、粒径分布和溶血磷脂酰胆碱等,结果表明中试工艺制备的丙泊酚脂肪乳注射液在强光照射、低温循环条件下稳定,溶血磷脂酰胆碱对温度比较敏感,6个月加速试验和24个月长期试验表明丙泊酚脂肪乳注射液各项质量指标符合要求。
