微孢子虫在线虫中的感染情况研究

【摘要】:模式生物秀丽隐杆线虫C.elegans 被广泛应用于多种实验室研究中,但对野生C.elegans的生态学研究十分匮乏。本文以寄生虫微孢子虫为研究重点,首先通过分子的方法,利用特定基因片段的扩增和系统发育进化树的分析,对41个野生线虫株系进行了微孢子虫感染鉴定。研究结果确定了 Nenatocida parisii是野生线虫C.elegans最常见的微孢子虫寄生虫;在微孢子虫门中,新发现并命名了 六个新 Nemmatocida 种,两个新属 Enteropsectra 和 Pancytospora 以及四个新种。其次,利用微分干涉显微镜和透射电子显微镜等技术手段,对可感染线虫的微孢子虫进行了形态学的观察和比较。再次,利用实验室感染实验,研究了可感染线虫的微孢子虫的寄主特异性。论文的另一部分以寄主C.elegans为主要研究对象,利用转基因C.elegans株系研究比较了 C.elegabs对不同微孢子虫感染的转录反应;通过食物消耗速率实验,研究比较了不同野生C.elegans株系对同种微孢子虫的敏感性差异,并进一步用荧光原位杂交技术研究比较了感染率和感染程度;通过生存竞争实验,研究了不同野生C.elegans株系对微孢子虫的敏感性差异可能导致的自然选择的进化后果。
[Abstract]:C. elegans, a model organism, has been widely used in many laboratory studies, but the ecological studies of wild C. elegans are scarce. Based on the amplification of specific gene fragments and phylogenetic tree analysis, 41 wild nematode strains were identified for microsporidium infection. The results showed that Nenatocida parisii was the most common microsporidium parasite of C. elegans. Six new Nemmatocida species, two new genera Enteropsectra and Pancytospora and four new species were discovered and named in the phylum of microsporidium. Secondly, differential interference microscope and transmission electron microscope were used. The morphology of microsporidium infected with nematodes was observed and compared. Thirdly, the host specificity of infected nematode was studied by laboratory infection experiment. The other part of the paper mainly studied the host C. elegans. The transcriptional response of C. elegans to different microsporidium infection was compared by using transgenic C. elegans strain, and the sensitivity of C. elegans strains to microsporidium homogeneae was compared by food consumption rate experiment. The infection rate and the degree of infection were compared by fluorescence in situ hybridization, and the evolutionary consequences of natural selection due to the different sensitivity of different wild C. elegans strains to microsporidium were studied by survival competition experiment.
