内源干扰物和中药小分子对斑马鱼生长发育的影响

【摘要】:内源干扰物对生长发育的影响还没有得到充分的研究,然而早期接触内源干扰物可能会在之后的发育中产生永久有害的影响。转基因斑马鱼的组织由于结合了荧光蛋白而成为检测给定物质有无影响的强大工具。我们利用7种转基因株系检测了6种己知的内源干扰物和3种药物对于斑马鱼生长发育的影响。检测表明其中4种物质对于斑马鱼的四个不同器官组织生长发育产生了影响。内源干扰物四溴双酚A和3种种药物扰乱了斑马鱼血管系统的正常发育。其中组织蛋白去乙酰酶抑制剂曲古菌素A和丙戊酸还抑制了斑马鱼内分泌胰腺,外分泌胰腺,肝脏和咽头齿的生长发育。实验结果表明内源干扰物影响了斑马鱼的生长发育。斑马鱼转基因株系是活体快速检测内源干扰物对胚胎生长发育影响的良好模型。传统中药是现代中药的重要组成部分。然而对于传统中药及其有效成分对于成长发育影响的研究还比较少。我们利用斑马鱼对三种常见中药及其活性成分进行了测试。研究表明木通和黄芪脱水溶液对延迟了斑马鱼胚胎的生长发育,丹参脱水溶液及其三种主要活性物质单独或混合溶液对斑马鱼的血管系统的生长发育有不同程度的抑制作用。研究结果表明斑马鱼是进行小物质筛选的良好模型。实验结果也我们将内源干扰物和中药活性成为联系在一起,比如他们可能会有相似的作用目标,核受体。
[Abstract]:The effects of endogenous interference on growth and development have not been fully studied, however, early exposure to endogenous interference may have a permanent harmful effect in subsequent development. Genetically modified zebrafish tissues are powerful tools for detecting the effects of a given substance by combining fluorescent proteins. The effects of 6 known endogenous disruptors and 3 drugs on the growth and development of zebrafish were studied using 7 transgenic lines. The results showed that four of them affected the growth and development of four different organs of zebrafish. Intrinsically, tetrabromobisphenol A and 3 drugs disrupt the normal development of the zebrafish vascular system. Among them, histone deacetylase inhibitor Tracogenin A and valproic acid also inhibited the growth and development of the endocrine pancreas, exocrine pancreas, liver and pharyngeal head teeth of zebrafish. The results showed that the endogenous interference affected the growth and development of zebrafish. Zebrafish transgenic lines are a good model for rapid detection of the effects of endogenous interference on embryo growth and development in vivo. Traditional Chinese medicine is an important part of modern Chinese medicine. However, there are few studies on the effects of traditional Chinese medicine and its active components on growth and development. Three common Chinese medicines and their active components were tested by zebrafish. The results showed that Astragalus and Astragali dehydrated solution delayed the growth and development of zebrafish embryos, and the dehydration solution of Salvia miltiorrhiza and its three main active substances alone or mixed had different inhibitory effects on the growth and development of the vascular system of zebrafish. The results show that zebrafish is a good model for screening small substances. The results also linked endogenous disruptions to the activity of traditional Chinese medicine, for example, they might have similar targets, nuclear receptors.
