【摘要】:本论文包括两部分实践部分和实验部分。论文第一部分为中日友好医院药学部实践部分。分别是门诊中药房、住院中药房、中药库、中药制剂室、药检室。在这5个部门实践过程中,学习和了解了各部门的工作流程及注意事项。论文第二部分为是山海螺的化学成分与质量控制研究。主要分为三个部分,一、山海螺药材的研究进展;二、山海螺的化学成分研究;三、山海螺的质量控制研究。第一部分对山海螺的化学成分、药理作用、临床应用等方面进行论述。阐述了山海螺的化学成分主要为三萜皂苷类化合物,此外,还含有糖类、甾醇、生物碱类、黄酮类及挥发油等化合物。具有抗癌、抗突变、抗氧化、降血糖、降血脂、镇痛等作用。第二部分为山海螺的化学成分研究,采用HP-20大孔树脂、Sephadex LH-20柱色谱等色谱分离方法对山海螺50%乙醇洗脱部位进行化合物分离纯化,通过1 H-NMR和13 C-NMR等波谱技术和化学方法鉴定所分离纯化得到的化合物结构。通过反复上SephadexLH-20柱色谱共分离得到6个化合物,鉴定了 5个化合物,分别是5-羟甲基-2-呋喃甲醛(1)、CodonopilodiynosideA(2)、lobetyol(3)、(+)-isolariciresinol(4)、党参炔苷(5)。其中化合物(1)、(2)、(3)、(4)均为首次从该植物中分得。第三部分为山海螺的质量控制研究,运用HPLC建立党参炔苷含量测定的方法学,同时共收集了 13批山海螺药材进行HPLC测定,测得色谱图,运用中国药典委员会"中药色谱指纹图谱相似度评价软件A"进行评价,生成对照图谱。该方法操作简便快捷,具有专属性强、稳定性、重复性良好等特点,可用于山海螺药材的质量控制与评价。
[Abstract]:This paper includes two parts of practice and experiment. The first part of the thesis is the practical part of the pharmacy department of China-Japan Friendship Hospital. They are the pharmacy in the outpatient department, the pharmacy in the hospital, the Chinese medicine library, the Chinese medicine preparation room and the medicine inspection room. During the practice of these 5 departments, we learn and understand the work flow and attention of various departments. The second part of the thesis is to study the chemical constituents and quality control of Codonopsis lanceolata. Mainly divided into three parts. The research progress of medicinal materials, a study on the chemical constituents of Codonopsis lanceolata; two, the research on the quality control of Codonopsis lanceolata; three, Codonopsis lanceolata. The first part of Codonopsis lanceolata chemical composition, pharmacological effects and clinical application were discussed. Describes the three main chemical composition of Codonopsis lanceolata saponins compounds, in addition, also contains carbohydrates, sterols, alkaloids, flavonoids and volatile oil compounds. It has the effect of anticancer, anti - mutation, antioxidation, hypoglycemic, antilipidemia and analgesic effect. The second part is the study on the chemical constituents of Codonopsis lanceolata, using the separation method of HP-20 macroporous resin column chromatography, Sephadex LH-20 chromatography for separation and purification of compounds of 50% ethanol elution fraction of Codonopsis lanceolata, structure of pure compounds obtained by the separation of 1 H-NMR and 13 C-NMR spectra and chemical identification methods. 6 compounds were isolated by repeated SephadexLH-20 column chromatography. 5 compounds were identified, namely, 5- hydroxymethyl -2- furan formaldehyde (1), CodonopilodiynosideA (2), lobetyol (3), (+) -isolariciresinol (4), and Codonopsis glucoside (5). The compounds (1), (2), (3) and (4) were all derived from the plant for the first time. The third part is the research on the quality control of Codonopsis lanceolata, using HPLC to establish a method for determination of Lobetyolin content, and collected a total of 13 batches of medicinal materials of Codonopsis lanceolata HPLC were measured using Chinese chromatogram, Pharmacopoeia Commission "chromatographic fingerprint similarity evaluation software A to evaluate the generated control map. This method is simple and quick, has the characteristics of strong specificity, stability, good repeatability, and can be used for quality control and evaluation of medicinal materials of Codonopsis lanceolata.
