【摘要】:背景:目前,心血管疾病严重的威胁着人类健康,已经成为人类健康的头号杀手;稳定型心绞痛是一种常见的心血管疾病,它主要由心肌灌注与心肌需求平衡失调引起,其病理基础几乎都是冠状动脉粥样硬化性狭窄;在过去十年中,发明并使用了各种靶向治疗药物用来治疗心血管疾病,但是在患者生存率方面并未得到相应的提高。近几年来,为了进一步降低心血管相关疾病的死亡率,科学家们对非编码RNA(non-coding RNA,nc RNA)与心血管相关疾病之间的联系进行了艰苦卓绝的探索与研究,比如长链非编码RNA(long noncoding RNA,lnc RNA)和短链非编码微RNA(micro RNA,mi RNA)。Lnc RNA是含有超过200个核苷酸,能够调节基因表达的内源性RNA。最近的研究提示lnc RNAs在心血管疾病的发生和发展过程中起着关键作用。例如,Bvrt,Fendrr,ANRIL,MIAT,LIPCAR和My Heart(Mhrt)在心血管发育、心脏疾病、心肌梗死、心肌病、心力衰竭和动脉粥样硬化中起着重要作用。此外,最新的研究表明在许多病理状态的发生和发展中,lnc RNAs的表达失调;这提示lnc RNA可能可以成为理想的治疗靶点和生物标志物。然而,lnc RNA作为稳定型心绞痛的生物标志物目前没有任何相关的研究报道。在前期的lnc RNA微阵列研究中,我们发现稳定型组中lnc RNA ENST00000589524.1的表达明显增高。目的:本研究通过测定ENST00000589524.1在稳定型心绞痛患者及健康人群中的表达水平,评估ENST00000589524.1是否可作为稳定型心绞痛的生物标志物,并分析ENST00000589524.1表达水平与稳定型心绞痛患者的相关危险因素。材料和方法:选取38例稳定型心绞痛(stable angina pectoris)患者及38例健康对照组,均来自2014年11月至2015年6月期间相继在新疆医科大学第一附属医院心血管内科住院的患者。采用q RT-PCR方法去测定稳定型心绞痛患者和健康对照组患者中ENST00000589524.1的表达水平。结果:q RT-PCR分析结果显示ENST00000589524.1的表达水平在稳定型心绞痛患者中呈上调趋势。在本研究中,我们没有发现lnc RNA表达水平与稳定型心绞痛相关风险因素。结论:ENST00000589524.1是稳定型心绞痛的潜在生物标志物。
[Abstract]:Background: at present, cardiovascular disease is a serious threat to human health, has become the number one killer of human health; stable angina is a common cardiovascular disease, which is mainly composed of myocardial perfusion and myocardial demand imbalance caused by the pathological basis of almost all coronary atherosclerotic stenosis; in the past ten years, and invention the use of a variety of targeted drugs used in the treatment of cardiovascular disease, but did not get the corresponding increase in the rate of survival. In recent years, in order to further reduce cardiovascular disease mortality, scientists of non encoding RNA (non-coding RNA, NC RNA) to explore and research with extremely hard and bitter cardiovascular diseases for example, a long chain of non encoding RNA (long noncoding RNA, LNC RNA) and short chain non micro RNA (micro encoding RNA, MI RNA).Lnc RNA contains more than 200 nucleotides, A recent study of endogenous RNA. can regulate gene expression suggesting that LNC RNAs plays a key role in the occurrence and development of cardiovascular diseases. For example, Bvrt, Fendrr, ANRIL, MIAT, LIPCAR and My Heart (Mhrt) in the development of cardiovascular, heart disease, myocardial infarction, heart disease, heart failure and plays an important role in atherosclerosis in addition, the latest research shows that the occurrence and development in many pathological conditions, disorder of expression of LNC RNAs; it is suggested that LNC RNA may be an ideal therapeutic targets and biomarkers. However, LNC RNA as a biomarker for stable angina. There are no related research reports. LNC RNA microarray study in the early stage, we found that the expression of LNC RNA ENST00000589524.1 in the stable group was significantly increased. Objective: This study through the determination of ENST00000589524.1 in patients with stable angina pectoris and The expression level of healthy people, to assess whether ENST00000589524.1 can be used as stable angina biomarkers, and analyze the risk in patients with ENST00000589524.1 expression and stable angina. Methods: 38 patients with stable angina pectoris (stable angina pectoris) patients and 38 healthy controls were from November 2014. To June 2015 have in patients with cardiovascular diseases hospitalized in the First Affiliated Hospital of Xinjiang Medical University. To determine stable angina patients and healthy control group the expression level of ENST00000589524.1 in patients with Q RT-PCR method. Results: Q RT-PCR analysis showed that the expression levels of ENST00000589524.1 were up-regulated in patients with angina pectoris in stable type. In this study, we found no LNC RNA expression level in patients with stable angina related risk factors. Conclusion: ENST00000589524. 1 is a potential biomarker of stable angina pectoris.
