【摘要】:癌症是威胁人类健康的一大杀手,其中宫颈癌是威胁女性的第二大恶性疾病。研究数据表明,对于早期癌症的治疗效果明显优于中晚期癌症。这就意味着如果能在癌症初期对其进行准确的诊断,就能改善病人的生存质量甚至可以延长病人的生存年限。对于癌症的早期诊断,目前主要的方法是基于免疫组织化学、分子生物学、流式细胞术等检测肿瘤标记物的病理学检测方法。但这些方法都存在着假阳性和假阴性的问题。原子力显微镜技术由于具有纳米及微纳米级分辨率以及对细胞无任何损伤的特点而得到了广泛的应用。本论文通过这项技术,检测宫颈癌细胞的性状变化,进而分析出细胞内部的病理变化,达到癌细胞早期诊断的效果。同时,为了探索宫颈癌的治疗办法,本文进一步研究了经抗癌药多西他赛处理后的宫颈癌细胞的性状变化。本文利用原子力显微镜在相同的测量条件下分别对药物处理前后的正常宫颈上皮细胞和宫颈癌HeLa细胞进行了扫描,并对两者的力曲线进行分析与比较,研究了细胞的物理化学性质变化。通过分析得知细胞经过药物处理后细胞的物理化学性质发生了改变,细胞的塑性指数发生变化,说明细胞的内部结构有所改变,这一改变通过利用Western Blot检测细胞微管蛋白的含量变化得到证明;细胞的粘滞功发生改变,细胞表面的弹性模量发生变化,这一改变通过透射电镜的观察得到证明,通过透射电镜的观察我们知道细胞表面的微绒毛经过药物处理后显著减少;细胞的弹性等物理化学性质也投相应的改变。通过AFM测得的实验结果都通过相关生物学实验进一步验证了原子力显微镜对癌细胞诊断结果的正确性。原子力显微镜对细胞的这种无标记检测手段弥补了这一领域的空白,使得它在未来的科学研究中能起到至关重要的作用。此外,原子力显微镜也有望成为医学检测方向的重要检测工具。
[Abstract]:Cancer is a major threat to human health, including cervical cancer is the second malignant diseases threatening women. Research data show that in late stage of early cancer treatment is superior in cancer. This means that if you can make accurate diagnosis of the quality of life in the early stage of cancer, can improve the patients can even be extended the patient's survival. For the early diagnosis of cancer, at present, the main method is based on immunohistochemistry, molecular biology, pathology detection method of flow cytometry detection of tumor markers. But these methods exist in the problem of false positives and false negatives. Atomic force microscopy has been widely used because of the characteristics of nano and micro nano resolution of cells without any damage. This paper through this technology, detection of changes of characteristics of cervical cancer cells, and then analyze the fine The pathological changes inside the cell, to achieve the effect of cancer cells in early diagnosis of cervical cancer. At the same time, in order to explore the way of treatment, this paper further studies the changes of characteristics of anticancer drugs docetaxel after treatment of cervical cancer cells. By using atomic force microscopy in the measurement under the same circumstances respectively before and after drug treatment of normal cervical epithelial cells and HeLa cervical cancer cells were scanned, and the stress curves are analyzed and compared, studied the change of physical and chemical properties of cells. Cells that treated cells after the physical and chemical properties changed through analysis, cell plasticity index changes, shows the internal structure of cells changed, the a change has been proved by changing the content of cells was detected by Western Blot tubulin; viscous power cells changed, the elastic modulus of the cell surface occurs Change, this change was demonstrated by transmission electron microscopy, transmission electron microscope and we know that the microvilli were significantly reduced after treated by medicine; physical and chemical properties of the elastic cell also cast the corresponding change. Measured by AFM experimental results through biological experiments further verified by atomic force microscopy the diagnosis results of cancer cells. The atomic force microscope for cell label free detection means to make up for gaps in this field, so that it can play a crucial role in scientific research in the future. In addition, an important tool for detection of atomic force microscope is expected to become the direction of medical testing.
