【摘要】:目的:我们拟寻找与无功能型垂体腺瘤的侵袭性相关的重要蛋白,为全面、系统研究无功能型垂体腺瘤侵袭生长的机制提供理论依据,为今后利用蛋白质组学方法全面、系统研究无功能型垂体腺瘤的病因,以及分子靶向治疗、基因治疗提供新的突破点。方法:收集2015年10月至2016年10月期间新疆维吾尔自治区人民医院神经外科收治的经本院手术治疗的18例无功能型垂体瘤病人的肿瘤标本(侵袭性生长9例,非侵袭性生长9例)。用二维凝胶电泳结合质谱技术寻找在无功能型垂体腺瘤中表达的差异性蛋白;应用双向凝胶电泳(2-DE)结合基质辅助激光解析电离飞行时间质谱技术(MALDI-TOF-MS)对侵袭性无功能型垂体腺瘤及非侵袭性无功能型垂体腺瘤肿瘤组织进行蛋白质翻译水平的筛选,并比较组织蛋白的异同,寻找与无功能型垂体腺瘤侵袭性相关的蛋白质及可能在临床诊断中应用的蛋白质。结果:(1)通过双向电泳结果(通过PDQuest软件显不同组的差异表达蛋白质点),建立了无功能型垂体腺瘤的的2-DE图谱。(2)通过研究我们找到4种差异蛋白点,其中p14ARF蛋白在侵袭性无功能型垂体腺瘤的肿瘤组织中较非侵袭性无功能型垂体腺瘤的肿瘤表达下调,EGFR、HMGA2、PIK3CA蛋白在侵袭性无功能型垂体腺瘤的肿瘤组织中较非侵袭性无功能型垂体腺瘤的肿瘤表达上调。结论:这4种差异蛋白可能与无功能型垂体腺瘤的侵袭性生长有关。
[Abstract]:Objective: to search for important proteins related to invasiveness of non-functional pituitary adenomas, and to provide theoretical basis for a comprehensive and systematic study of the mechanism of invasiveness and growth of non-functional pituitary adenomas. In order to study the etiology and molecular targeted therapy of non-functional pituitary adenoma systematically by proteomics. Gene therapy provides a new breakthrough. Methods:. From October 2015 to October 2016, 18 patients with disfunctional pituitary adenoma treated in neurosurgery department of Xinjiang Uygur Autonomous region people's Hospital were collected (. Invasive growth was found in 9 cases. Two dimensional gel electrophoresis and mass spectrometry were used to search for differentially expressed proteins in non-functional pituitary adenomas. Two-dimensional gel electrophoresis (2-DE) combined with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MSM). The protein translation level of invasive and non-invasive pituitary adenomas was screened. The differences and similarities of histone were compared. Search for proteins associated with invasiveness of nonfunctional pituitary adenomas and proteins that may be used in clinical diagnosis. PDQuest software was used to show the differentially expressed protein spots in different groups. The 2-DE map of non-functional pituitary adenoma was established. The expression of p14ARF protein was down-regulated in tumor tissues of invasive non-functional pituitary adenomas than that of non-invasive pituitary adenomas. The expression of PIK3CA protein was up-regulated in invasive non-functional pituitary adenomas than in non-invasive pituitary adenomas. Conclusion:. These four differentially expressed proteins may be related to the invasive growth of nonfunctional pituitary adenomas.
