泡型肝包虫病患者外周血Treg细胞的水平变化及其临床意义

【摘要】:目的:通过流式细胞技术比较分析泡型肝包虫病患者与健康人之间外周血CD4~+CD25~+CD127~(low)/Fox P3~+Treg细胞水平的差异,观察他们之间的关系,探讨其临床意义。方法:选择青海大学附属医院2016.8--2017.01期间住院患者25例作为实验组,健康体检者25例作为对照组。实验组在术前和术后10天分别采集外周血标本,通过流式细胞技术比较分析泡型肝包虫病患者与健康人之间外周血CD4~+CD25~+CD127~(low)/Fox P3~+Treg细胞水平的差异,分析他们之间的关系。结果:CD4~+CD25~+CD127~(low) Treg细胞水平:正常人(3.19±0.33)和患者术前(12.95±1.13)相比有统计学差异(P0.05),且患者术前的外周血CD4~+CD25~+CD127~(low)Treg细胞水平较高;正常人(3.19±0.33)和患者术后(5.70±1.21)相比有统计学差异(P0.05),且患者术后的外周血CD4~+CD25~+CD127~(low) Treg细胞水平较高;患者术前(12.95±1.13)和患者术后(5.70±1.21)相比有统计学差异(P0.05),且患者术前的外周血CD4~+CD25~+CD127~(low) Treg细胞水平较高。CD4~+CD25~+Fox P3~+Treg细胞水平:正常人(1.85±0.50)和患者术前(4.99±0.98)相比有统计学差异(P0.05),且患者术前的外周血CD4~+CD25~+Fox P3~+Treg细胞水平较高;正常人(1.85±0.50)和患者术后(2.84±0.57)相比有统计学差异(P0.05),且患者术后外周血CD4~+CD25~+Fox P3~+Treg细胞水平较高;患者术前(4.99±0.98)和患者术后(2.84±0.57)相比有统计学差异(P0.05),且患者术前的CD4~+CD25~+Fox P3~+Treg细胞水平较高。正常人的外周血CD4~+CD25~+CD127~(low) Treg细胞和CD4~+CD25~+Fox P3~+Treg细胞有相关性(r=0.568,P0.05),且两指标呈正相关。结论:1.泡型包虫患者CD4~+CD25~+CD127~(low)/Fox P3~+Treg细胞水平均显著升高,可能与患者免疫抑制状态形成有关。2.泡型包虫病患者外周血CD4~+CD25~+CD127~(low)/Fox P3~+Treg细胞水平术前显著高于手术后,提示根治性手术可能能够影响患者体内免疫状态。3.据前两点可知,外周血Treg细胞水平可在一定程度上反应治疗效果、监测病情复发。
[Abstract]:Objective: to compare and analyze CD4 ~ CD25 ~ CD127 of peripheral blood between patients with alveolar hepatic hydatid disease and healthy persons by flow cytometry. The difference of cell level of / / Fox P3 ~ Treg. Methods: 25 inpatients from the affiliated Hospital of Qinghai University from June 8 to January 2017.01 were selected as the experimental group. 25 healthy persons were used as control group. Peripheral blood samples were collected before and 10 days after operation in the experimental group. Comparative analysis of peripheral blood CD4 ~ CD25 ~ CD127 low between patients with alveolar hepatic hydatid disease and healthy persons by flow cytometry. The difference of cell level of / / Fox P3 ~ Treg. The relationship between them was analyzed. Results the Treg cell levels of CD4 ~ CD25 ~ CD127) in normal subjects were 3.19 卤0.33) and those in patients before operation (3. 19 卤0. 33). 12.95 卤1.13) compared with the statistical difference (P0.05). The level of CD4 ~ CD25 ~ CD127~(low)Treg cells in peripheral blood was higher than that in patients before operation. There was significant difference between normal subjects (3.19 卤0.33) and postoperative patients (5.70 卤1.21) (P 0.05). The level of CD4 ~ CD25 ~ CD127 Treg cells in peripheral blood after operation was higher. There was significant difference between preoperative and postoperative patients (12.95 卤1.13) and postoperative (5.70 卤1.21) (P 0.05). CD4 ~ CD25 ~ Fox P3 ~ Treg cell level was 1.85 卤0.50 in normal subjects and 4.99 卤0.98 in patients before operation. There was statistical difference (P0.05). The level of CD4 ~ CD25 ~ Fox P3 ~ Treg cells in peripheral blood was higher than that in patients before operation. There was significant difference between normal persons (1.85 卤0.50) and postoperative patients (2.84 卤0.57) (P 0.05). The level of CD4 ~ CD25 ~ Fox P3 ~ Treg cells in peripheral blood was higher after operation. There was significant difference between preoperative and postoperative patients (4.99 卤0.98) and postoperative (2.84 卤0.57) (P 0.05). The levels of CD4 ~ CD25 ~ Fox P3 ~ Treg cells were higher in patients before operation. Treg cells and CD4 ~ CD25 ~ Fox P3 ~ Treg cells were associated with rrmc0.568. P0.05). Conclusion the level of CD4 ~ CD25 ~ CD127~(low)/Fox P3 ~ Treg cells in patients with hydatid alveolar type increased significantly. 2. Peripheral blood CD4 ~ CD25 ~ CD127 of patients with alveolar hydatid disease may be related to the formation of immunosuppressive state in patients with hydatid disease. The cell level of / / Fox P3 ~ Treg was significantly higher before operation than that after operation. The results suggest that radical operation may affect the immune state of patients. According to the first two points, the level of peripheral blood Treg cells can respond to the therapeutic effect to a certain extent and monitor the recurrence of the disease.
