七氟醚对大鼠股动脉血栓形成的影响

【摘要】:目的:研究七氟醚对大鼠股动脉血栓后的影响及其作用机制。方法:选用体重为250~300g的SD大鼠40只(雌雄各半),随机分为4组:对照组(C组)、模型组(M组)、七氟醚低浓度组(SL组)和七氟醚高浓度组(SH组)。除了对照组以外,其余3组都用三氯化铁建立股动脉血栓模型,SL组和SH组经口持续吸入浓度分别为3%和4%的七氟醚两小时,对照组和模型组则持续吸入空气2h。大鼠心尖取血后处死。本实验测定大鼠血栓重量;HE染色法观察大鼠股动脉的病理变化;采用ELISA法测定肝脏,血浆中PF4、vWF、AT-Ⅲ、P-选择素、BCL-2以及BAX水平;采用蛋白印迹法检测大鼠股动脉BCL-2和BAX蛋白的表达水平。结果:(1)血栓重量称重结果:与模型组比较,用药组的血栓重量都减轻,具有差异性(P0.05或P0.01)。(2)HE染色结果:对照组股动脉的管腔内无血栓,血管内皮完整;模型组股动脉管腔内可见血栓充盈,血管内皮剥脱严重,七氟醚低浓度组血管堵塞较多,血管内皮剥脱较重,七氟醚高浓度组血管堵塞较轻,血管内皮损伤轻微。(3)ELISA:[1]肝脏指标检测结果:1.与对照组比较,模型组PF4和vWF含量均升高(均P0.05);与模型组比较,用药组的PF4和vWF含量明显下降(P0.05)。2.与对照组比较,模型组P-选择素和AT-Ⅲ含量均升高(均P0.05);与模型组比较,七氟醚低、高剂量组的P选择素和AT-Ⅲ含量明显下降(P0.05,P0.01)。3.和对照组比较,模型组BAX和BCL-2含量均升高(P0.01);与模型组比较,七氟醚低、高剂量组的BCL-2含量明显下降(P0.05),而BAX含量明显下降(P0.01)。[2]血浆指标检测结果:1.与对照组比较,模型组PF4和vWF含量均升高(均P0.05);和模型组比较,七氟醚低、高剂量组的PF4和vWF含量明显下降(P0.05)。2.与对照组比较,模型组的BAX明显升高,BCL-2含量明显降低(均P0.01);与模型组比较,七氟醚低、高剂量组的BAX含量明显下降,BCL-2含量明显升高(均P0.05)。[3]BCL-2和BAX的蛋白表达水平的变化:与对照组比较,模型组股动脉BAX蛋白表达量显著增加;与模型组比较,七氟醚不同浓度组BCL-2明显升高,BAX明显下降,(均P0.01),BCL-2/BAX比值明显升高。结论:(1)七氟醚对动脉血栓的形成有抑制作用。(2)七氟醚抑制血栓形成的机制可能是通过保护血管内皮细胞、防止血小板活化以及抑制细胞凋亡来实现的。
[Abstract]:Objective: to study the effect and mechanism of sevoflurane on femoral artery thrombosis in rats. They were randomly divided into 4 groups: control group (C), model group (M), sevoflurane low concentration group (SL group) and sevoflurane high concentration group (SH group), except for control group. The other three groups were treated with ferric chloride to establish femoral artery thrombosis model. Sevoflurane concentrations of 3% and 4% sevoflurane were continuously inhaled by oral administration in SL and SH groups for two hours, respectively. The rats in the control group and the model group were killed after inhalation of air for 2 hours. The weight of thrombus was measured in this experiment. The pathological changes of femoral artery in rats were observed by HE staining. The levels of P- selectin BCL-2 and BAX in liver and plasma were determined by ELISA. The expression of BCL-2 and BAX protein in femoral artery of rats was detected by Western blot. Results: compared with the model group, the weight of thrombus was reduced in the drug group. The results showed that there was no thrombus in the femoral artery and the vascular endothelium was intact in the control group. In the model group, thrombosis could be seen in femoral artery lumen, vascular endothelium exfoliation was serious, vascular clogging was more in low concentration group of sevoflurane, vascular endothelium exfoliation was more serious, vascular clogging was lighter in high concentration group of sevoflurane.
