喉鳞状细胞癌中脂肪酸代谢相关基因ADRP和ACAA1异常表达的研究

【摘要】:研究背景喉癌是头颈部最常见的恶性肿瘤之一,其中绝大多数为喉鳞状细胞癌(Laryngeal squamous cell carcinoma,LSCC),近年来其发病率呈上升趋势,其发病机制尚不甚清楚,因此学者们致力于探索喉癌发生发展机制。肿瘤细胞能量代谢改变如Warburg效应在肿瘤增殖、调亡、转移等方面发挥重要作用。脂肪酸是能量代谢的重要底物,肿瘤细胞中脂肪酸代谢呈异常激活状态,脂肪酸代谢相关基因表达水平的改变可能影响肿瘤细胞的生物学行为。研究目的为进一步探讨喉鳞状细胞癌中脂肪酸代谢机制,我们开展本课题研究即脂肪酸代谢相关基因脂肪相关分化蛋白(Adipose differentiation-related protein,ADRP)和乙酰辅酶A酰基转移酶(Acetyl CoA acyltransferase 1,ACAA1)基因在LSCC中的表达特点,分析其表达与临床病理参数的关系,为进一步的实验研究提供理论基础。研究方法1.通过实时荧光定量PCR方法检测LSCC中的ADRP和ACAA1基因的转录水平。2.通过用免疫组织化学染色法从蛋白水平检测LSCC中ADRP和ACAA1的异常表达。3.结合LSCC样本的临床特征,分析脂肪酸代谢基因ADRP和ACAA1与喉癌发生发展的关系。研究结果1.检测发现相对于癌旁组织,ADRP和ACAA1在LSCC中转录水平下调。2.LSCC中ADRP和ACAA1的蛋白水平呈明显表达下调。3.ADRP和ACAA1的表达与LSCC的TNM分期、分化程度有关。研究结论脂肪酸代谢相关基因ADRP和ACAA1在LSCC中异常表达,此外与LSCC的TNM分期、病理分级有关,提示了ADRP和ACAA1的异常表达可能参与LSCC的发生发展过程,为探索LSCC的发生机制提供新的思路。
[Abstract]:Background laryngeal carcinoma is one of the most common malignant tumors in the head and neck, most of which are laryngeal squamous cell carcinomas. Therefore, scholars have been working to explore the mechanism of laryngeal cancer. Changes in energy metabolism of tumor cells, such as Warburg effect, play an important role in tumor proliferation, apoptosis, metastasis and so on. Fatty acids are important substrates of energy metabolism. Fatty acid metabolism is abnormally activated in tumor cells. The changes of fatty acid metabolism related gene expression may affect the biological behavior of tumor cells. The purpose of this study was to further explore the mechanism of fatty acid metabolism in laryngeal squamous cell carcinoma (LSCC). In this study, we studied the expression of adipose differentiation-related protein (ADRP) and Acetyl CoA acyltransferase 1 (ACAA1) gene in LSCC, and analyzed the relationship between the expression and clinicopathologic parameters. To provide a theoretical basis for further experimental research. 1. Detection of the transcription level of ADRP and ACAA1 genes in LSCC by real-time fluorescent quantitative PCR. 2. Detection of ADRP in LSCC by immunohistochemical staining from protein level. And abnormal expression of ACAA1. 3. Combined with the clinical characteristics of LSCC samples, To analyze the relationship between fatty acid metabolism gene ADRP and ACAA1 and the occurrence and development of laryngeal carcinoma. 1. The expression of ADRP and ACAA1 protein was down-regulated in LSCC compared with that of adRP and ACAA1 in paracancerous tissue. 2. The expression of ADRP and ACAA1 in LSCC was down-regulated. The expression of ACAA1 was associated with the TNM staging of LSCC. Conclusion the abnormal expression of fatty acid metabolism-related genes ADRP and ACAA1 in LSCC is related to the TNM stage and pathological grade of LSCC, suggesting that the abnormal expression of ADRP and ACAA1 may be involved in the occurrence and development of LSCC. It provides a new idea for exploring the mechanism of LSCC.
