杜仲叶的品质评价研究

【摘要】:目的:研究并比较不同产地杜仲叶药材的品质、四种化学成分的定性定量分析、微量元素的含量测定,为杜仲叶药材的品质进行评价。方法:根据2015版《中华人民共和国药典》和查阅大量文献对杜仲叶药材进行性状鉴定;对杜仲叶药材粉末进行显微鉴别;对7批杜仲叶药材中化学成分绿原酸进行薄层鉴别;使用紫外分光光度计测定7批杜仲叶提取液中的总多糖、总黄酮、总多酚含量;采用傅里叶变换红外光谱法(FTIR)对杜仲叶药材进行红外光谱的研究,快速建立杜仲叶药材红外指纹图谱,并对7批杜仲叶进行聚类分析;利用高效液相色谱法(HPLC)测定7批杜仲叶药材中绿原酸、槲皮素、山奈酚和芦丁的含量;采用火焰原子吸收分光光度法测定7批杜仲叶药材、土壤样品、灌溉水样品中钾、钠、钙、镁、铁、锌、锰7种金属元素的含量,采用石墨炉原子吸收法测定土壤样品中铅的含量和7批杜仲叶药材、土壤样品、灌溉水样中铜的含量。结果:薄层色谱鉴别最优固定相选择青岛海洋H高效板,以乙酸丁酯-甲酸-水(7:2.5:2.5)的上层溶液为展开剂,7批杜仲叶薄层色谱中所含成分基本一致;7批杜仲叶药材提取液中总多糖含量在16.02-37.87 mg/g之间、总黄酮含量在30.97-119.07 mg/g之间、总多酚含量在9.89-36.74 mg/g之间,7批杜仲叶药材总成分的含量均有所差异,其中,样品S7的总多糖、总黄酮、总多酚含量均为较高;从杜仲叶药材的红外光谱峰形、峰位等信息可知,7批杜仲叶光谱特征基本一致,在通过二阶导数变换后,使得7批药材红外光谱之间的差异放大,通过聚类分析对杜仲叶进行分类,可将杜仲叶药材分为5组;7批杜仲叶药材中绿原酸、槲皮素、山奈酚、芦丁的含量有所差异,其中绿原酸含量在0.8353-3.3624 mg/g之间、槲皮素含量在0.1608-0.6129 mg/g之间、山奈酚含量在0.0236-0.1031 mg/g之间、芦丁含量在0.0022-0.0043 mg/g之间;7批杜仲叶药材中钾、钠、钙、镁、铁、锌、锰、铜的含量分别在15300.95-40300.95、58.24-298.98、3319.15-9515.96、8335.17-12138.92、211.40-933.63、21.92-72.62、144.79-560.05、209.21-406.58μg/g之间,土壤样品中钾、钠、钙、镁、铁、锌、锰、铜、铅的含量平均含量为43310.19、678.89、9015.96、511.03、22836.26、118.53、656.90、47.93、21.69μg/g之间,灌溉水样品中钾、钠、钙、镁、铁、锌、锰、铜的含量平均含量为534.72、7065.13、2840.43、2086.00、38.71、0.13、0.186、18.16μg/mL之间,其中钾、钙等元素含量普遍较高,重金属元素检查均符合国家标准。结论:本研究初步探索并比较新疆产杜仲叶和其他4个产地杜仲叶中总多糖、总黄酮、总多酚含量;对7批药材进行了性状鉴定、显微鉴别、薄层鉴别;采用HPLC法测定7批杜仲叶药材中绿原酸、槲皮素、山奈酚、芦丁的含量;测定了杜仲叶药材、土壤样品、灌溉水样品中微量元素的含量。为建立科学的杜仲叶品质标准提供科学依据,使新疆产杜仲叶药用资源得到广泛的开发和利用。
[Abstract]:Objective: to study and compare the quality of Eucommia ulmoides leaves from different areas, the qualitative and quantitative analysis of four chemical constituents, and the determination of trace elements. Methods: according to the 2015 edition of the Pharmacopoeia of the people's Republic of China and consulting a large number of literature to identify the character of Eucommia ulmoides leaves, to identify the powder of Eucommia ulmoides leaves by microscope, to evaluate the quality of Eucommia ulmoides L. The content of total polysaccharides, total flavonoids and total polyphenols in 7 batches of Eucommia ulmoides leaves extract was determined by UV spectrophotometer. Fourier transform infrared spectroscopy (FTIR) was used to study the infrared spectrum of Eucommia ulmoides leaves. The infrared fingerprint of Eucommia ulmoides leaves was established quickly, and 7 batches of Eucommia ulmoides leaves were analyzed by cluster analysis. The contents of Lv Yuan, quercetin, kaempferol and rutin in 7 batches of Eucommia ulmoides leaves were determined by HPLC, and potassium and sodium in 7 batches of Eucommia ulmoides leaves, soil samples and irrigation water samples were determined by flame atomic absorption spectrophotometry. The contents of calcium, magnesium, iron, zinc and manganese in soil samples were determined by graphite furnace atomic absorption spectrometry (GFAAS). The content of Copper in Irrigation Water samples. Results: TLC was used to identify the optimal stationary phase in Qingdao Ocean H High efficiency Plate. The contents of total polysaccharides and flavonoids were between 16.02-37.87 mg/g and 30.97-119.07 mg/g in TLC of 7 batches of Eucommia ulmoides leaves by TLC with butyl acetate-formic acid-water 7: 2.5: 2.5) as the developing agent, and the contents of total polysaccharides in the extracts of Eucommia ulmoides ulmoides were between 16.02-37.87 mg/g and 30.97-119.07 mg/g, respectively. The contents of total polyphenols in the leaves of Eucommia ulmoides were all different between 9.89-36.74 mg/g, and the contents of total polysaccharides, total flavonoids and total polyphenols of sample S7 were higher. The spectral characteristics of Eucommia ulmoides leaves in 7 batches of Eucommia ulmoides were basically the same. After the second derivative transformation, the infrared spectra of 7 batches of Eucommia ulmoides were amplified, and the leaves of Eucommia ulmoides were classified by cluster analysis. The contents of Lv Yuan, quercetin, kaempferol and rutin in the leaves of Eucommia ulmoides ulmoides could be divided into 5 groups. The contents of Lv Yuan acid, quercetin and kaempferol were 0.8353-3.3624 mg/g, 0.1608-0.6129 mg/g and 0.0236-0.1031 mg/g, respectively. The contents of potassium, sodium, calcium, calcium, magnesium, iron, zinc, manganese and copper in seven batches of Eucommia ulmoides leaves were between 0.0022 and 0.0043 mg/g. The average contents of potassium, sodium, calcium, magnesium, iron, manganese, copper and copper were 15300.95-40300.95n 58.24-298.98.93319.15-9515.915.919.15-9515.95.12138.911.40-933.6321.92-72.6144.569-0.05209.21-406.58 渭 g / g of potassium, sodium, calcium, magnesium, iron, zinc, manganese, copper and lead, respectively. Conclusion: the contents of total polysaccharides, total flavonoids and total polyphenols in leaves of Eucommia ulmoides from Xinjiang and other four areas were preliminarily explored and compared. The content of Lv Yuan, quercetin, kaempferol and rutin in 7 batches of Eucommia ulmoides leaves were determined by HPLC. The content of trace elements in irrigation water samples provides a scientific basis for establishing scientific quality standards of Eucommia ulmoides leaves and makes the medicinal resources of Eucommia ulmoides leaves widely developed and utilized in Xinjiang.
