压力合并高脂饮食对动脉粥样硬化的影响及越鞠丸的干预作用

【摘要】:目的:为了观察压力对小鼠血清炎症反应以及动脉粥样硬化斑块的稳定性的影响,从炎症之角度探讨压力在动脉粥样硬化发生发展中所起的作用,并予越鞠丸进行干预,观察中医药在压力致动脉粥样硬化发生中的干预作用。方法:实验通过予以不同应激刺激建立慢性应激的动物模型,将48只Apo E-/-小鼠随机分为6组,空白组、模型组、越鞠丸低倍剂量+阿托伐他汀组、越鞠丸中倍剂量+阿托伐他汀组、越鞠丸高倍剂量+阿托伐他汀组、阿托伐他汀组,六组均予高脂饮食,除外空白组其余各组随机安排应激实验持续十二周,实验开始即给药,十二周后采血查E选择素、hs-CRP,取主动脉应用免疫组化方法分析神经肽Y、E选择素、脂蛋白磷脂酶A2在小鼠主动脉中的表达差异并计算主动脉内斑块的易损指数。另设一组取正常8只C57B1/6的雄性小鼠,正常饮食喂养,不予应激试验,不予药物治疗。结果:予压力刺激的小鼠血清中hs-CRP、E-选则素的含量增加,标志着炎症反应的发生,予越鞠丸干预各组的主动脉中E选择素、神经肽Y、脂蛋白磷脂酶A2的表达较模型组减少,E选择素中可见越鞠丸的剂量效应。同时以易损指数评估,炎症反应的发生与斑块的稳定性呈负相关。结论:压力可以增加冠心病发病的风险,是冠心病的危险因素之一。压力下的小鼠血清hsCRP、E-选则素的含量增加,表明慢性压力应激可导致炎症反应的发生;同时通过病理学检测也证明,予慢性压力应激刺激后小鼠主动脉中E选择素、神经肽Y、脂蛋白磷脂酶A2的表达增多,炎症与斑块的稳定性呈负相关;予越鞠丸干预后,炎症标志物的含量下降,提示越鞠丸可在一定程度上缓解斑块炎症从而达到稳定斑块的作用,为从郁论治冠心病提供了有力证据。
[Abstract]:Objective: to observe the effect of pressure on the inflammatory response in serum and the stability of atherosclerotic plaques in mice, to explore the role of pressure in the occurrence and development of atherosclerosis from the perspective of inflammation, and to intervene with Yueju pill.To observe the effect of traditional Chinese medicine (TCM) on pressure-induced atherosclerosis.Methods: 48 Apo E-r-mice were randomly divided into 6 groups, blank group, model group and low-dose Atto vastatin group.The middle dose Atto vastatin group, the high dose Atto vastatin group, the Atto vastatin group, the six groups were given high-fat diet, except the blank group, the other groups were randomly assigned to the stress test for 12 weeks. The drug was given immediately at the beginning of the experiment.After 12 weeks, blood samples were collected to examine the expression of E-selectin hs-CRP.The expression of neuropeptide Ya-E selectin and lipoprotein phospholipase A _ 2 in mouse aorta was analyzed by immunohistochemical method and the vulnerability index of plaques in aorta was calculated.Another group of 8 normal C57B1/6 male mice were fed with normal diet, no stress test, no drug treatment.Results: the content of hs-CRPnE- selectin in the blood serum of pressure-stimulated mice was increased, indicating the occurrence of inflammatory reaction. Yueju pill was given to interfere with E-selectin in aorta of each group.Compared with the model group, the expression of neuropeptide Y, lipoprotein phospholipase A2 decreased the dose-effect of Yueju pill.At the same time, the occurrence of inflammation and plaque stability were negatively correlated with vulnerability index.Conclusion: stress can increase the risk of coronary heart disease and is one of the risk factors of coronary heart disease.The increase of serum hsCRP- E- selectin in mice under stress suggests that chronic stress can lead to inflammatory reaction, and that E selectin in aorta of mice stimulated by chronic stress is also proved by pathological examination.The expression of neuropeptide Y, lipoprotein phospholipase A2 was increased, and inflammation was negatively correlated with plaque stability.The results suggest that Yueju pill can alleviate plaque inflammation and stabilize plaque to a certain extent, which provides strong evidence for treating coronary heart disease from depression.
