ATF5在鼻咽癌侵袭和转移中的作用及其机制研究

目的:探讨转录激活因子5(Activating transcription factor 5,ATF5)对鼻咽癌细胞侵袭及迁移能力的影响及其可能分子机制。方法:以人鼻咽癌CNE2Z细胞为研究对象,采用慢病毒载体shRNA-ATF5干扰技术沉默CNE2Z细胞ATF5表达及实时荧光定量聚合酶链反应(Quantitative Real-time Polymerase Chain Reaction,qRT-PCR)检测慢病毒shRNA-ATF5载体干扰效率;以未感染慢病毒的CNE2Z细胞作空白对照(Control组),慢病毒空载体LV-shRNA-NC感染CNE2Z细胞作为阴性对照(NC组),慢病毒载体shRNA-ATF5(LV-shRNA-ATF5)感染CNE2Z细胞为实验组(shRNA-ATF5组);再运用噻唑蓝(Methylthiazolyl Blue Tetrazolium,MTT)法及克隆形成实验检测CNE2Z细胞增殖能力,划痕实验、Transwell迁移实验、侵袭实验检测CNE2Z细胞迁移及侵袭能力;采用Affymetrix人类基因表达谱芯片技术比较感染shRNA-ATF5慢病毒后...
