高含油樟树无性系内生菌多样性及筛选

目前对樟树(Cinnamomum camphora(L.)Presl.)的研究主要集中在精油成分分析、扦插育苗技术和矮化造林等方面,尚未深入微生物层面分析,尤其探索樟树内生菌分离、鉴定及与宿主植物及次生产物生长发育之间的联系更是尚未涉猎。因此,本研究采用16srDNA高通量测序技术对三个季节的样品的内生菌进行测序,研究樟树在不同季节(生长期)的生理生化、内生菌群落多样性及次生代谢,同时筛选出促生树樟精油的优势内生菌,为研究樟树内生菌的功能及其应用提供理论基础。本研究通过对5个樟树无性系叶片的生理特性、生化指标和精油含量及精油成分来探究内生菌与樟树的生理生化及精油(含量和成分)的相关性。并且采用组织块分离法,主要通过显微镜观察,分离纯化樟树内生细菌和真菌。使用分子鉴定策略进一步分析不同无性系内生菌多样性、代表性细菌和真菌。通过固相微萃取纯化内生菌之后,提取和分离的代谢产物,测定代谢产物成分,并与樟树叶精油成分进行比对,来筛选促进樟树叶精油含量的内生菌。获得的结果如下。在不同的采样时间,获得了来自叶子的10,844,124个有效序列。Shannon指数表明,在采样期间,细菌群落的多样性较低...
