左归丸对D-半乳糖致老化小鼠模型认知功能和听功能损伤的干预作用

【摘要】:目的观察左归丸对D-半乳糖致老化小鼠认知功能和听功能的影响。方法将雄性C57BL/6小鼠随机分为4组,模型组、左归丸低剂量和高剂量组按照200 mg/kg皮下注射D-半乳糖,对照组注射等体积生理盐水,连续造模60 d。左归丸低剂量和高剂量组在造模的第41天开始按2 g/kg和4 g/kg的剂量连续灌胃左归丸混悬液20 d。采用Morris水迷宫观察小鼠的空间学习和记忆能力,采用听性脑干反应(ABR)检测小鼠听功能。结果 Morris水迷宫定位航行实验结果显示,从训练的第3天开始,模型组的潜伏期明显高于对照组,左归丸低剂量和高剂量组的潜伏期均低于模型组。空间探索实验结果显示,模型组穿越平台次数和目标象限时间百分比明显低于对照组(P0.01,P0.05);左归丸低剂量组和高剂量组的穿越平台次数和目标象限百分比均高于模型组,其中高剂量组与模型组比较差异有统计学意义(P0.05)。ABR结果显示,在8 k Hz和16 k Hz时模型组听阈值明显高于对照组,左归丸低剂量组和高剂量组听阈值均低于模型组(P0.05,P0.01)。结论左归丸对D-半乳糖导致的听功能损伤和认知功能下降有保护作用。
[Abstract]:Objective To observe the effect of Zuogui Pill on D- galactose induced aging mice cognitive function and auditory function effects. Methods male C57BL/6 mice were randomly divided into 4 groups, model group, Zuoguiwan group with low dose and high dose 200 mg/kg subcutaneous injection of D- galactose, the control group were given normal saline, spatial learning and memory the ability of continuous model 60 D. Zuoguiwan low dose and high dose group made in the forty-first day at 2 g/kg and 4 g/kg dose by intragastric administration of Zuogui pill suspension 20 D. mice were observed by Morris water maze, the auditory brainstem response (ABR) test in mouse auditory function. Results Morris water maze navigation experiment results show that from the beginning of training for third days, the model group was significantly higher than the control group, Zuoguiwan low dose and high dose group were significantly lower than in the model group. The experimental results show that the exploration of space, time and number of crossing platform model group The percentage of target quadrant was significantly lower than the control group (P0.01, P0.05); Zuogui pill low dose group and high dose group the number of crossing platform and target quadrant percentage were higher than that of model group, which was statistically significant difference between high dose group and model group (P0.05).ABR results showed that in 8 K Hz and 16 K Hz when listening to the threshold of model group was significantly higher than the control group, Zuogui pill low dose group and high dose group thresholds were lower than the model group (P0.05, P0.01). Conclusion Zuogui Pill on D- galactose induced damage of auditory function and cognitive decline has a protective effect.
