日本血吸虫感染ICR小鼠后Tc1和Tc2细胞的动态观察及其功能分析

【摘要】:目的观察ICR小鼠感染日本血吸虫后不同时期T淋巴细胞中杀伤性T细胞(Tc)亚型Tc1和Tc2比例的动态变化,并分析两者分别与Th1和Th2细胞比例变化的相关性。方法分别取日本血吸虫感染0、3、5、8、13周的ICR小鼠脾脏淋巴细胞,采用流式细胞术检测各个时期Tc1、Tc2和Th1、Th2细胞亚群分别占T淋巴细胞的比例。体外用SWA及SEA刺激小鼠脾脏淋巴细胞48h后,,采用流式细胞术检测脾淋巴细胞中Tc1、Tc2细胞以及Th1、Th2细胞分别占T淋巴细胞的比例。结果与0周(未感染)对照比较,日本血吸虫感染3、5、8、13周的小鼠T淋巴细胞中Tc1细胞比例显著增高(P均0.01),感染5、8、13周的小鼠T淋巴细胞中Tc2细胞比例显著增高(P均0.01)。比较Tc1和Tc2细胞比例在相同时间点的增速发现,感染血吸虫后的前3周Tc1细胞比例增高最显著,而感染3-5周内Tc2细胞比例的增高最显著。血吸虫感染后各个时间点CD3~+T细胞中的Th1细胞比例与Tc1细胞比例平行增长,且两者呈正相关(r=0.978,P=0.004);CD3~+T细胞中的Th2细胞比例与Tc2细胞比例平行增长,且两者呈正相关(r=0.974,P=0.005)。体外实验表明可溶性成虫抗原(SWA)刺激脾细胞可优势增加T淋巴细胞中的Tc1细胞比例(P 0.01),而可溶性虫卵抗原(SEA)刺激脾细胞可优势增加T淋巴细胞中的Tc2细胞比例(P 0.01)。结论日本血吸虫感染不同阶段,小鼠T淋巴细胞中的Tc1和Tc2细胞比例均显著升高,并分别与Th1和Th2细胞比例变化呈正相关,根据两者的增速,提示Tc1和Tc2分别可能促进了Th1和Th2的分化。SWA可优势诱导Tc1细胞,而SEA可优势诱导Tc2细胞。
[Abstract]:Objective to observe the dynamic changes of Tc1 and Tc2 subtypes in T lymphocytes of ICR mice at different stages after infection with Schistosoma japonicum (Schistosoma japonicum). Methods the spleen lymphocytes of ICR mice infected with Schistosoma japonicum were collected for 813 weeks after infection with Schistosoma japonicum. The percentage of Tc1Tc2 and Th1Th2 cell subsets in T lymphocytes was detected by flow cytometry in each stage. After stimulation with SWA and SEA for 48 h in vitro, the ratio of Tc1Tc2 and Th1Th2 cells to T lymphocytes was measured. The percentage of Tc1tc2 cells and Th1Th 2 cells in splenic lymphocytes were detected by flow cytometry. The results were compared with those of 0 weeks (uninfected). The proportion of Tc1 cells in T lymphocytes of mice infected with Schistosoma japonicum for 813 weeks was significantly higher than that in mice infected with Schistosoma japonicum. The proportion of Tc2 cells in T lymphocytes of 13 weeks old mice increased significantly (P < 0.01). Comparing the growth rate of Tc1 and Tc2 cells at the same time, we found that the ratio of Tc1 cells to Tc2 cells increased at the same time. The proportion of Tc1 cells increased most significantly in the first three weeks after infection with Schistosoma japonicum. The proportion of Th1 cells in CD3 ~ T cells increased parallel to that of Tc1 cells at different time points after infection of Schistosoma japonicum. There was a positive correlation between them. The proportion of Th2 cells in CD3 ~ T cells increased in parallel with the proportion of Tc2 cells, and there was a positive correlation between them. In vitro experiments showed that SWA-induced splenocytes could increase the proportion of Tc1 cells in T lymphocytes (P 0.01). But soluble egg antigen (SEAA) can increase the proportion of Tc2 cells in T lymphocytes by stimulating spleen cells. Conclusion Schistosoma japonicum infection in different stages. The percentage of Tc1 and Tc2 cells in T lymphocytes of mice increased significantly, and was positively correlated with the changes of Th1 and Th2 cells, respectively, according to the growth rate of the two cells. These results suggested that Tc1 and Tc2 could promote the differentiation of Th1 and Th2, respectively. SWA could induce Tc1 cells preferentially, while SEA could induce Tc2 cells preferentially.
