苏黄止咳胶囊对咳嗽变异性哮喘豚鼠模型疗效的实验研究

【摘要】：【目的】观察苏黄止咳胶囊对咳嗽变异性哮喘(cough variant asthma,CVA)豚鼠模型的疗效,探讨苏黄止咳胶囊的作用机制。【方法】将50只豚鼠随机分成正常对照组、模型对照组、醋酸泼尼松组及苏黄止咳胶囊高、低剂量组,复制CVA豚鼠模型,检测各组豚鼠咳嗽反应、肺组织病理学、支气管肺泡灌洗液(BALF)中嗜酸粒细胞百分率及血清中白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、Ig E的水平变化。【结果】与正常对照组比较,模型对照组豚鼠咳嗽次数显著增多(P0.05);与模型对照组比较,苏黄高剂量组咳嗽次数显著减少(P0.05)。与正常对照组比较,模型对照组BALF中嗜酸性粒细胞百分率显著上升(P0.05);3个治疗组嗜酸性粒细胞百分率均较模型组显著降低(P0.05)。治疗组肺组织病理改变较模型对照组减轻。与正常对照组比较,模型对照组血清中IL-4、TNF-α、Ig E水平显著升高(P0.05);与模型对照组比较,醋酸泼尼松组血清中IL-4、TNF-α水平显著下降,苏黄高剂量组血清中TNF-α、Ig E水平显著下降(P0.05)。【结论】苏黄止咳胶囊可能通过降低血清中TNF-α和Ig E的水平,改善CVA豚鼠的咳嗽症状、气道炎症及肺组织病理变化,从而达到治疗CVA的目的。
[Abstract]:[objective] to observe the effect of Suhuang Zhike capsule on cough variant asthmatic guinea pig model of cough variant asthma. To explore the mechanism of Suhuang Zhike capsule. [methods] 50 guinea pigs were randomly divided into normal control group, model control group, prednisone acetate group and Suhuang Zhike capsule high and low dose group, CVA guinea pig model was established. Cough reaction, lung histopathology, percentage of eosinophils in bronchoalveolar lavage fluid (BALF) and interleukin-4 (IL-4) in serum were measured in each group. The levels of IFN- 纬, TNF- 伪 and Ig E in IFN- 纬, TNF- 伪, TNF- 伪 were compared with those in normal control group. In the model control group, the number of cough increased significantly (P 0.05). Compared with the model control group, the cough frequency of Suhuang high dose group was significantly decreased than that of normal control group. In model control group, the percentage of eosinophil in BALF increased significantly (P 0.05). The percentage of eosinophilic granulocytes in the three treatment groups was significantly lower than that in the model group, and the pathological changes of lung tissue in the treatment group were less than those in the model control group. Serum IL-4 TNF- 伪 Ig E level in the model control group was significantly higher than that in the control group (P 0.05). Compared with the model control group, the serum levels of IL-4 and TNF- 伪 in prednisone acetate group decreased significantly, and TNF- 伪 in serum in the high dose group of Su Huang. [conclusion] Suhuang Zhike capsule may improve cough symptoms of CVA guinea pigs by lowering the levels of TNF- 伪 and IgE in serum. Airway inflammation and pathological changes of lung tissue, so as to achieve the purpose of treatment of CVA.
