针灸对环磷酰胺模型荷瘤小鼠骨髓细胞DNA修复相关蛋白动态调节的研究

【摘要】:目的:观察针灸对环磷酰胺(CTX)化疗后的荷瘤小鼠白细胞及骨髓细胞DNA修复蛋白O6-甲基鸟嘌呤-DNA甲基转移酶。(MGMT)、DNA聚合酶β(POLβ)、切除修复交叉互补基因(XPD)表达的影响。方法:160只雄性清洁级昆明种小鼠植种S180肉瘤,按体质量随机分为荷瘤空白(A)组、荷瘤模型(B)组、荷瘤针刺(C)组、荷瘤艾灸(D)组。B、C、D组腹腔注射CTX 150mg/kg制备CTX荷瘤模型小鼠,A组注射同等剂量的0.9%氯化钠溶液。C、D组分别选取"大椎""膈俞""肾俞""足三里"进行治疗,A、B组每日陪同抓取固定,不治疗。镜检各组小鼠外周血白细胞,免疫组化法检测骨髓细胞DNA修复蛋白MGMT、POLβ、XPD的表达情况,统计分析数据。结果:针灸可提升化疗后荷瘤小鼠白细胞数量(P0.05),亦能上调CTX化疗后荷瘤小鼠骨髓细胞DNA修复相关蛋白MGMT、POLβ、XPD的表达。结论:针灸升高CTX化疗后荷瘤小鼠外周血白细胞的疗效确切,其升白机制与上调CTX化疗后荷瘤小鼠骨髓细胞DNA修复相关蛋白MGMT、POLβ、XPD的表达及改善化疗后骨髓抑制关系密切。
[Abstract]:Objective: to observe the effect of acupuncture and moxibustion on DNA repair protein O6-methylguanine-DNA methyltransferase (O6-methylguanine-DNA methyltransferase) in leukocytes and bone marrow cells of tumor-bearing mice after chemotherapy with cyclophosphamide (CTX). DNA polymerase 尾 -POL 尾, excision and repair of cross complementary gene (XPD) expression. Methods S180 sarcoma was implanted in 1: 160 male Kunming mice of clean grade. According to body mass, they were randomly divided into three groups: group A, group B), group C (acupuncture), group D (moxibustion). Group D was intraperitoneally injected with CTX 150mg / kg to make CTX tumor bearing model mice. Group A was injected with the same dose of 0.9% sodium chloride solution. Group D selected "Dazhui", "Geshu", "Shenshu" and "Zusanli" respectively to carry out the treatment of AZA B group daily accompanied by grab fixation, no treatment. Microscopic examination of peripheral blood leukocytes of mice in each group. The expression of DNA repair protein (MGMT-POL 尾 -XPD) in bone marrow cells was detected by immunohistochemistry. Results: acupuncture and moxibustion could increase the number of leukocytes in tumor-bearing mice after chemotherapy and up-regulate the MGMT of DNA repair related protein in bone marrow cells of tumor-bearing mice after chemotherapy with CTX. Conclusion: acupuncture and moxibustion can increase the leucocyte expression in peripheral blood of tumor-bearing mice after CTX chemotherapy. The mechanism of leukopenia was closely related to the up-regulation of the expression of DNA repair related protein MGMT-POL 尾 -XPD in bone marrow cells of tumor-bearing mice after chemotherapy and the improvement of bone marrow suppression after chemotherapy.
