【摘要】:目的明确人偏肺病毒减毒突变株(human metapneumovirus attenuated strain ,M_2)的免疫原性及保护效果,评价其做为疫苗的应用前景,为hMPV减毒活疫苗的开发奠定坚实理论基础。 方法清洁级6~7周龄、雌性BALB/c小鼠144只随机分为3个组,分别为DMEM组、M_2组和WT组,每组48只。WT组每只小鼠鼻腔滴入10~7PFU/ml NL/1/00-gfp株hMPV病毒液100μl。M_2组每只小鼠鼻腔滴入10~7PFU/ml M_2减毒突变株病毒液100μl,DMEM组小鼠鼻腔滴入100μl DMEM培养基,滴鼻后第5、14、28、56天麻醉并处死每组动物各8只。3组分别在首次滴鼻后28、56天再次经鼻腔滴入10~7PFU/ml NL/1/00-gfp株hMPV病毒液100μl。并于再次滴鼻后第5天处死动物。每次每组各8只。均收集血清和肺组织用于检测血清中和抗体滴度、肺组织病毒滴度及病理改变。 结果M_2组与WT组相比,M_2毒力减弱,肺内复制时间缩短,但所诱导的中和抗体滴度水平与野生型相似,且对两亚型流行株具有交叉中和活性。平均抗NL/1/00血清抗体滴度水平高于平均抗NL/1/99。高效稳定的抗体水平产生完整的下呼吸道免疫保护作用。 结论M_2作为hMPV减毒活疫苗候选疫苗,具有很好的应用前景。
[Abstract]:Objective to identify the attenuated human metapneumovirus attenuated strain of human metapulmonary virus. The immunogenicity and protective effect of M _ 2) and its application prospect as vaccine were evaluated to lay a solid theoretical foundation for the development of live attenuated hMPV vaccine. Methods 144 female BALB/c mice of 6 and 7 weeks old were randomly divided into three groups: DMEM group (group 2) and group WT (group B). WT group (n = 48). WT group (n = 48) was dripped into the nasal cavity of 10 ~ 7 PFU / ml / mouse. The NL/1/00-gfp strain hMPV virus solution was 100 渭 l in the nasal cavity of each mouse in 10 ~ 7PFU / ml M2 attenuated mutants. The mice in DMEM group were anesthetized on day 514, 2856 after nasal drip with 100 渭 l DMEM medium, and 8 animals in each group were killed respectively 28 days after the first nasal drip. 56 days again through the nasal cavity into the 10 ~ (7) PFU / ml. NL/1/00-gfp strain hMPV virus solution was 100 渭 l. The animals were killed on the 5th day after nasal drip again. Each group had 8 animals. Serum and lung tissues were collected for detection of serum neutralizing antibody titer. Viral titer and pathological changes in lung tissue. Results compared with WT group, compared with WT group, the virulence of M _ 2 was decreased and the time of lung replication was shortened, but the titer of neutralizing antibody induced by M _ (2) was similar to that of wild type. The average antiserum antibody titer of NL/1/00 was higher than that of the average anti-NL / 1 / 99. The high and stable antibody level produced complete lower respiratory tract immunity protection. Function. Conclusion as a candidate vaccine for hMPV attenuated live vaccine, M _ (2) has a good prospect of application.
