【摘要】:目的分析产气肠杆菌携带blaNDM-1质粒的转移性、质粒复制子分型及blaNDM-1的基因环境。方法产气肠杆菌HN-NDM0711为研究对象,接合实验鉴定质粒转移性,PCR方法对质粒复制子分型,染色体步移技术对blaNDM-1上下游测序,基因组序列比对使用BLASTN和BLASTP,注释使用Vector NTI 11.5.1。结果产气肠杆菌HN-NDM0711接合实验阳性,质粒复制子为IncA/C型;blaNDM-1位于不常见的插入序列ISAba14和IS91之间;在blaNDM-1上游出现一个Tn3转座子和I型整合子,整合子上含有一个由庞大镶嵌序列构成的罕见耐药基因盒。结论 IncA/C型质粒pHN-NDM0711携带blaNDM-1及耐药基因盒来源于不同抗菌药物选择压力环境下的基因重组。只有严格控制临床、工业和农业抗菌药物的合理使用才能从源头上减少此类细菌的产生。
[Abstract]:Objective to analyze the transferability of Enterobacter aerogenes carrying blaNDM-1 plasmid. Methods Enterobacter aerogenes HN-NDM0711 was used as the research object. PCR method was used to type plasmid replicas, chromosome step technique was used to sequence blaNDM-1 upstream and downstream, and BLASTN and BLASTP were used for genome sequence alignment. Vector NTI 11.5.1.Results the HN-NDM0711 conjugation test of Enterobacter aerogenes was positive, and the plasmid replicon was IncA/C type. BlaNDM-1 is located between the unusual insert sequence ISAba14 and IS91; A Tn3 transposon and type I integron appeared in the upstream of blaNDM-1. The integron contains a rare drug resistant gene box composed of large mosaic sequences. Conclusion. IncA/C plasmid pHN-NDM0711 carries blaNDM-1 and drug resistance gene cassette from gene recombination under different antimicrobial selection pressure. The rational use of industrial and agricultural antimicrobial agents can reduce the production of such bacteria at the source.
