筋骨痛消丸对兔膝关节骨关节炎模型关节液IL-1、TNF-α水平的影响

【摘要】：目的:观察筋骨痛消丸干预后膝OA模型兔膝关节液中IL-1、TNF-α含量的变化,探讨筋骨痛消丸治疗膝骨性关节炎的作用机制。方法:将42只新西兰兔随机抽出11只纳入空白组,余纳入模型组,模型组用管状石膏伸直位固定右后膝5周制作膝骨性关节炎模型,完成造模后,随机抽取空白组和模型组各1只兔子处死以验证模型成功后,拆除造模组石膏。将没有造模的剩余10只兔子分为A组(空白组),造模成功的剩余30只兔子随机分为B组(模型组)、C组(筋骨痛消丸组)、D组(盐酸氨基葡萄糖组)。C组予以与60Kg体重人等效剂量的筋骨痛消丸悬浊液10mL、D组予以与60Kg体重人等效剂量的盐酸氨基葡萄糖混悬液10mL灌胃,A、B组予以10mL蒸馏水灌胃,灌胃处理3周后处死,行各组肉眼观察膝关节大体形态以及取股骨髁软骨行光镜观察,ELISA法测定膝关节液中IL-1以及TNF-α含量。结果:①造模5周后,造模组膝关节出现明显的退行性改变。②治疗后肉眼观察B、C、D组较A组相比均有退变,C、D组的退变程度较B组轻。光镜下B、C、D组均可见软骨退变的病理变化。③治疗后:B、C、D组与A组相比较关节液中IL-1和TNF-α含量明显升高(P0.05).C、D组关节液中IL-1和TNF-α含量与B组相比有明显减少(P0.05),治疗后C组IL-1、TNF-α含量与D组比较无显著差异(P0.05)。结论:筋骨痛消丸对兔膝骨性关节炎有防治作用,其机理可能是通过降低关节液中细胞炎性因子IL-1、TNF-α含量从而抑制软骨的破坏,延缓骨性关节炎的进展。
[Abstract]:Objective: the rabbit model of knee OA in knee joint fluid observation of Jingu Tongxiao pills after the intervention of IL-1, changes of TNF- alpha content, to explore the mechanism of action of Jingu Tongxiao pills in the treatment of knee osteoarthritis. Methods: 42 New Zealand rabbits were randomly selected 11 rats into control group, more than in model group, model group with tubular gypsum right knee unbend fixed after 5 weeks for knee osteoarthritis model, complete model, randomly selected control group and model group with 1 rats in each group rabbits were sacrificed to verify the model after the success of the model.
