【摘要】:目的 探讨导致自然流产的染色体异常类型和荧光原位杂交(fluorescence in situ hybridization, FISH)技术在检测自然流产组织染色体非整倍体畸变中的应用价值。 方法 本研究以107例自然流产组织作为研究对象,对全部标本进行细胞培养,并对细胞培养成功的标本进行染色体G显带核型分析;应用13、16、18、21、22、X和Y染色体区域特异性探针对107例自然流产组织进行FISH检测;对上述两种方法的检测结果比较分析。 结果 (1)107例标本培养成功76例,核型分析与FISH技术的可检测率分别为71.03%(76/107)、100%(107/107),两者差异有显著性(X2=36.25,P0.05); (2)107例自然流产标本异常率为46.73%,核型分析检测出异常35例,FISH检测出异常48例,两者检测结果一致率为96.05%; (3)染色体异常是导致自然流产的主要原因,主要异常类型是三体型(40.00%)、三倍体(28.00%)及X单体(20.00%)。 结论 (1) FISH技术与常规染色体核型分析检测结果一致; (2) FISH技术检测具有快速、准确的特点,同时由于FISH技术可分析大量的间期核,有利于嵌合体的诊断; (3)染色体异常是导致自然流产的重要原因。
[Abstract]:Purpose To investigate the types of chromosomal abnormalities and fluorescence in situ hybridization (fluorescence in situ hybridization) resulting in spontaneous abortion. Application value of fish technique in detecting chromosomal aneuploidy in spontaneous abortion. Method In this study, 107 cases of spontaneous abortion tissues were studied. All the samples were cultured and chromosome G-banding karyotype was analyzed. The FISH of 107 cases of spontaneous abortion tissues were detected by using the region specific probe of chromosome 1316, 1821, 22X and Y, respectively. The results of the two methods are compared and analyzed. Results 76 cases were successfully cultured. The detectable rates of karyotype analysis and FISH technique were 71.0333.776 / 107and 100 / 107 / 107, respectively. There was a significant difference between the two groups (P 0.05). The abnormal rate of 107 cases of spontaneous abortion was 46.73 and the karyotype analysis showed that 35 cases were abnormal and 48 cases were abnormal by fish. Chromosome abnormality is the main cause of spontaneous abortion. The main abnormal types are triploid 40.00m, triploid 28.00) and X monomer 20.00. Conclusion 1) the results of FISH were consistent with those of conventional chromosome karyotype analysis. (2) FISH detection is rapid and accurate, and because FISH can analyze a large number of interphase nuclei, it is helpful for the diagnosis of chimerism. Chromosome abnormality is an important cause of spontaneous abortion.
