补肾活血饮对帕金森病伴发抑郁模型大鼠行为学及脑内5-HT、NE及NPY的影响

【摘要】:目的探讨补肾活血饮对帕金森病伴发抑郁模型大鼠行为学及脑内单胺类神经递质5-羟色胺(5-HT)、去甲肾上腺素(NE)及神经肽Y(NPY)的影响。方法采用直接向脑内注入6-羟基多巴胺损毁脑黑质致密部的方法建立帕金森病模型,并采用慢性不可预见性温和应激及孤养方法建立抑郁模型。将造模成功的75只PD大鼠用随机数字表法分为模型组、补肾活血饮组、盐酸氟西汀组、多巴丝肼组、盐酸氟西汀+多巴丝肼组,每组15只;另取20只正常大鼠为正常组。通过糖水偏爱测试、强迫游泳实验、悬尾实验等方法检测大鼠行为;采用高效液相-电化学检测(HPLC-ECD)测定各组大鼠脑内5-HT、NE含量;用放射免疫法测定各组大鼠脑组织及血清中NPY含量。结果与正常组比较,给药前、治疗后模型组大鼠体重明显下降(P0.01);治疗后,模型组大鼠悬尾静止时间、强迫游泳不动时间均明显延长,糖水偏爱率明显减少,大鼠脑组织中5-HT及NE含量减少,血清及脑组织中NPY含量降低(均P0.01)。与模型组比较,治疗后补肾活血饮组、盐酸氟西汀组、盐酸氟西汀+多巴丝肼组大鼠体重升高(P0.01),悬尾静止时间、强迫游泳不动时间均减少(P0.01),糖水测试糖水偏爱率明显增高(P0.01),血清及脑组织中NPY含量升高(P0.01);补肾活血饮组、盐酸氟西汀组及盐酸氟西汀+多巴丝肼组、多巴丝肼组大鼠脑组织中5-HT及NE含量均升高(P0.01,P0.05)。与多巴丝肼组比较,补肾活血饮组、盐酸氟西汀组、盐酸氟西汀+多巴丝肼组大鼠体重升高(P0.01),悬尾静止时间、强迫游泳不动时间均减少(P0.05),糖水偏爱率增高(P0.05),血清及脑组织中NPY含量明显升高(P0.01)。结论补肾活血饮能促进PD伴发抑郁模型大鼠脑组织NPY释放,增加5-HT、NE含量,改善帕金森大鼠抑郁样行为。
[Abstract]:Objective to investigate the effects of Bushen Huoxue decoction (BHXY) on the behavior and brain monoamine neurotransmitter 5-hydroxytryptamine (5-HT) in rats with Parkinson's disease (PD) and depression. Methods Parkinson's disease model was established by direct injection of 6-hydroxydopamine into the brain to damage the substantia nigra compactness. The depression model was established by chronic unpredictable mild stress and solitary nourishment. 75 PD rats were randomly divided into three groups: model group, Bushen Huoxue Yin group and fluoxetine hydrochloride group. Dobutazid group, fluoxetine hydrochloride dobutazide group, 15 rats in each group; Another 20 normal rats were taken as normal group. The behavior of rats was detected by sugar water preference test, forced swimming test and tail suspension test. HPLC-ECD was used to detect the content of 5-HT NE in the brain of rats in each group by high performance liquid chromatography (HPLC) and electrochemical detection (HPLC-ECD). The contents of NPY in brain tissue and serum of rats in each group were measured by radioimmunoassay. After treatment, the rest time of tail suspension, the immobility time of forced swimming, the preference rate of sugar water and the contents of 5-HT and NE in brain tissue of the model group were significantly prolonged. The content of NPY in serum and brain tissue was lower than that in model group (P 0.01). Compared with model group, Bushen Huoxue Yin group and fluoxetine hydrochloride group after treatment were significantly lower than those in model group. In fluoxetine dobutazide group, the weight of rats increased P0.01, the rest time of tail suspension and the immobility time of forced swimming decreased P0.01). Sugar water test showed that the preference rate of sugar water was significantly higher than that of the control group (P 0.01), and the content of NPY in serum and brain tissue was higher than that in the control group (P 0.01). The contents of 5-HT and NE in brain tissue of rats in Bushen Huoxue Yin group, fluoxetine hydrochloride group, fluoxetine hydrochloride dobutazid group and dobutazid group were all increased (P0.01). Compared with dobutazid group, Bushen Huoxue decoction group, fluoxetine hydrochloride group, fluoxetine hydrochloride group increased body weight and suspended tail rest time. The immobility time of forced swimming decreased P0.05, and the preference rate of sucrose water increased P0.05). Conclusion Bushen Huoxue decoction can promote the release of NPY and increase the content of 5-HT NE in brain tissue of PD rats with depression. To improve the depressive behavior of Parkinson's rats.
