刚地弓形虫垂直传播小鼠模型的建立

【摘要】:目的探讨建立刚地弓形虫稳定垂直传播小鼠模型的方法。方法将42只孕8dBALB/c小鼠随机分为(A-F组),正常对照A组(孕鼠经腹腔注射无菌PBS 0.2ml/只)和感染组(B~F)(每只小鼠经腹腔接种不同剂量的弓形虫速殖子10、20、40、80个/0.2ml)。观察并记录孕鼠的体重改变、行为反应、流产率与活胎率;孕18d时剖腹,取出胎鼠进行形态学观察。结果感染组小鼠出现倦怠、竖毛、弓背等异常反应,且随速殖子感染数量的增加异常反应程度逐渐严重;F组孕鼠死亡2只。感染组较对照组体重增加不明显,F组体重减轻;孕18d时D~F组体重与对照组比较差异有统计学意义(P0.05)。感染组出现流产、死胎等,F组存活孕鼠腹中无胎鼠。D~F组流产率与对照组相比差异有统计学意义(P0.05);B~F组活胎率与对照组比较差异有统计学意义(P0.05)。E~F组胎鼠体重、体长、尾长与对照组比较差异有统计学意义(P0.05)。结论成功建立刚地弓形虫垂直传播小鼠模型,BALB/c小鼠孕8d腹腔注射接种RH株弓形虫速殖子40个/(0.2ml·只)为适宜感染剂量。
[Abstract]:Objective to establish a stable vertical transmission model of Toxoplasma gondii (Toxoplasma gondii). Normal control group A (pregnant rats injected aseptic PBS 0.2ml / mouse) and infected group (each mouse intraperitoneally inoculated with Toxoplasma gondii Toxoplasma Toxoplasma Toxoplasma Tachyzoites 1020M40). 80 / 0.2ml. the body weight change, behavioral reaction, abortion rate and live birth rate of pregnant rats were observed and recorded. On the 18th day of gestation, the fetal mice were taken out for morphological observation. Results in the infected group, abnormal reactions such as burnout, bristles and back of the bow were observed, and the abnormal reaction degree was gradually serious with the increase of tachyzoite infection. Two pregnant rats died in group F. the weight loss of infection group was not significant compared with that of control group. At the 18th day of gestation, there was a significant difference in body weight between DU F group and control group (P 0.05). Abortion and stillbirth occurred in infection group. The abortion rate of group F was significantly higher than that of control group (P 0.05). Compared with the control group, there was a significant difference in the rate of live fetus in group B and F (P 0.05). The body length and body weight of fetal rats in group E were significantly higher than those in group B (P 0.05). Conclusion the vertical transmission model of Toxoplasma gondii was established successfully. BALB/c mice were injected intraperitoneally with RH strain Toxoplasma gondii Toxoplasma gondii Toxoplasma gondii Toxoplasma gondii 40 / 0. 2ml 路mouse) on the 8th day of gestation.
