结核分枝杆菌Rv3245蛋白抗原免疫学特性的研究

【摘要】:目的:研究结核分枝杆菌Rv3425蛋白免疫学特性,评估其诊断应用价值及在结核致病性中的作用。方法:将Rv3425基因克隆至p ET28a载体中,诱导表达Rv3425融合蛋白并进行纯化;利用ELISA、Western blot分析其抗原性与特异性;流式检测该抗原对巨噬细胞凋亡的影响。结果:获得了可溶性原核表达融合蛋白Rv3425,Rv3425蛋白能刺激灭活结核分枝杆菌免疫小鼠脾细胞产生高水平的特异性IFN-γ、纯化的Rv3425蛋白能与结核感染小鼠血清特异性结合,其特异性血清抗体Ig G及Ig M在结核病人中的水平明显高于健康人,发现该蛋白能诱导巨噬细胞的凋亡。结论:本研究发现结核分枝杆菌Rv3425蛋白具有较强的抗原性且能促进巨噬细胞的凋亡,这些发现对于结核病的诊断及致病机制的研究具有重要价值。
[Abstract]:Objective: to study the immunological characteristics of Rv3425 protein of Mycobacterium tuberculosis and to evaluate its diagnostic value and its role in the pathogenicity of tuberculosis. Methods: the Rv3425 gene was cloned into p ET28a vector to express and purify the Rv3425 fusion protein. The antigenicity and specificity of EISAL were analyzed by Western blot. Results: the soluble prokaryotic expression fusion protein Rv3425rv3425 could stimulate the production of high specific IFN- 纬 by inactivated Mycobacterium tuberculosis immunized mice spleen cells and purified Rv3425. The protein can specifically bind to the serum of mice infected with tuberculosis. The levels of IgG and IgM in tuberculosis patients were significantly higher than those in healthy persons. Conclusion: the Rv3425 protein of Mycobacterium tuberculosis has strong antigenicity and can promote the apoptosis of macrophages. These findings are of great value in the diagnosis and pathogenesis of tuberculosis.
