牡丹皮、赤芍与白芍对急性血瘀模型大鼠活血功效的比较研究

【摘要】:目的观察牡丹皮、赤芍与白芍对急性血瘀模型大鼠的影响,揭示三者在功效方面的异同,并探讨牡丹皮、赤芍与白芍活血功效的机制。方法大鼠sc盐酸肾上腺素结合冰水浸泡制作急性血瘀模型,以丹参注射液为阳性对照,观察牡丹皮、赤芍与白芍对血液流变学、凝血功能指标、血小板系统、血清中血栓素B2(TXB2)、6-酮-前列腺素(6-keto-PGF1α)的影响。结果与模型组相比,牡丹皮能够延长急性血瘀模型大鼠的凝血酶原时间(PT)、凝血酶时间(TT)(P0.05),显著抑制血小板黏附(P0.01),且上调血小板内皮型一氧化氮合酶(e NOS)与Ser1177位磷酸化的e NOS(p-e NOS)蛋白表达水平(P0.01、0.001),减少血清中TXB2(P0.05)及增加6-keto-PGF1α的量(P0.01);赤芍能够显著改善不同切变率下血瘀模型大鼠的全血黏度,延长部分活化凝血活酶时间(APTT)、PT、TT(P0.05、0.01),降低血小板黏附率(P0.05),减少血清中TXB2的量(P0.01)、增加6-keto-PGF1α的量(P0.01),上调血小板e NOS、p-e NOS蛋白表达水平(P0.05);白芍则对血瘀模型大鼠的全血黏度的影响并不显著,但能够显著延长APTT、TT(P0.05),降低血小板黏附率(P0.05),增加血清中6-keto-PGF1α的量(P0.05),同时显著增加血小板e NOS、p-e NOS蛋白表达水平(P0.05)。结论牡丹皮、赤芍和白芍均具有不同程度改善血液流变性、凝血系统、抑制血小板黏附率、增加血小板e NOS、p-e NOS蛋白表达水平,但牡丹皮、赤芍较白芍活血化瘀作用强。
[Abstract]:Objective to observe the effects of peony peel, red peony root and white peony root on acute blood stasis model rats, to reveal the similarities and differences among them, and to explore the effect of peony peel. Methods the acute blood stasis model was made in rats with Sc hydrochloric acid and ice water immersion. The peony skin, paeoniae rubra and Paeoniae Alba were used as positive control to observe the indexes of hemorheology and coagulation function of Paeonia rubra and Radix Paeoniae Alba and Radix Paeoniae Alba and Radix Paeoniae Alba. The effects of thromboxane B _ 2H _ 2T _ XB _ (2) -6-keto-PGF _ (1 伪) on the platelet system were compared with those in the model group. Peony skin can prolong the prothrombin time and thrombin time of acute blood stasis model rats. It can significantly inhibit the platelet adhesion P0.01C, and up-regulate the expression of platelet endothelial nitric oxide synthase (Enos) and Ser1177 phosphorylated e NOS(p-e NOSprotein. The whole blood viscosity of rats with blood stasis model was significantly improved by P0.01P _ (0.01) and the increase of 6-keto-PGF _ (1 伪) in serum by reducing serum TXB _ (2) P _ (0.05) and increasing 6-keto-PGF _ (1 伪), and the whole blood viscosity of rats with blood stasis model was significantly improved by Paeonia lanceolata at different shear rates. Prolonging partial activated thromboplastin time (P0.05), decreasing platelet adhesion rate (P0.05N), reducing the amount of TXB2 in serum (P0.01A), increasing the amount of 6-keto-PGF1 伪 (P0.01A), upregulating the expression level of platelet E-NOSS-p-e NOS protein (P0.05), while Paeonia lanceolata had no significant effect on the whole blood viscosity of rats with blood stasis model. But it can significantly prolong the level of P0.05, decrease the adhesion rate of P0.05, increase the amount of 6-keto-PGF _ 1 伪 in serum and increase the expression level of E-NOSP-e NOS protein. Conclusion Peony peony, Radix Paeoniae rubra and Radix Paeoniae Alba can improve the hemorheology and coagulation system in different degrees. The platelet adhesion rate was inhibited and the expression level of eNOSP-e NOS was increased, but peony peel and red peony had stronger effect on promoting blood circulation and removing blood stasis than Paeonia lanceolata.
