益生菌L.casei Zhang对大鼠内毒素性肝损伤保护作用的研究

【摘要】:本文旨在研究益生菌L.casei Zhang对内毒素性肝损伤大鼠的保护作用。造模方法:禁食12小时后,对大鼠腹腔注射脂多糖(LPS:50mg/kg)和D-半乳糖胺(D-GalN:300mg/kg)。(1)将大鼠分为四组:正常对照组A(n=16);模型组B(n=16),灌服生理盐水1m1/d,30d后造模;预防组C(n=20),灌服益生菌Lcasei Zhang1×109CFU/d,30d后造模;地塞米松组D(n=10),腹腔注射地塞米松、LPS和D-GalN,观察15d内大鼠的存活率,以此制备生存曲线;(2)取48只大鼠分为CTL、L/G、L/G+LCZ. L/G+DXM4组,分别作如下处理:CTL组(正常对照组)正常喂养、L/G组(模型组)灌胃生理盐水1m1/d,L/G+LCZ组(预防组)灌胃益生菌菌粉悬液2×109CFU/d, L/G+DXM组(地塞米松组)腹腔注射地塞米松(10m/gkg),各组饲喂基础饲料,31d后禁食12h, B、C、D三组腹腔注射LPS(50mg/kg)和D-GalN(300mg/kg),处理后16小时处死大鼠,测血浆丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)及肝脏组织中肿瘤坏死因子-a(TNF-a)、Toll受体4(TLR4)、过氧化物酶体增殖物激活受体-γ(PPARγ)、NO等细胞因子的含量,同时,测定肝脏组织中磷酸化核因子抑制蛋白(p-IκB)、PPARY、TLR4等蛋白的表达情况,此外,还观察肝脏切片病理学变化。结果发现:(1)生存曲线中,C组的生存率低于A组和D组,但明显高于B组,说明益生菌L.casei Zhang可以在一定程度上保护大鼠肝脏,减轻肝脏损伤对大鼠机体造成的损害;(2)L/G+LCZ组和L/G+DXM组的ALT、AST及肝脏组织中TNF-a、NO、TLR4、p-IκB的表达量显著低于L/G组(P0.05),L/G+LCZ组PPARy表达高于L/GL/G组(P0.05),;L/G+LCZ组和L/G+DXM组肝脏MDA含量显著低于L/G组(P0.05),SOD活力显著高于L/G组(P0.05);病理切片结果发现,益生菌干预治疗可缓减内毒素引起的急性肝细胞损伤。因此,益生菌L.casei Zhang对内毒素性肝损伤的治疗具有良好的保护作用,可用于功能性食品的开发。
[Abstract]:The aim of this study was to study the protective effect of probiotic L.casei Zhang on endotoxin induced liver injury in rats. Rats were divided into four groups by intraperitoneal injection of lipopolysaccharide (LPS: 50 mg / kg) and D-galactosamine (D-GalN: 300mg 路kg 路kg -1). In the dexamethasone group, the lipopolysaccharide (LPS) and D-GalN were injected intraperitoneally to observe the survival rate of rats within 15 days, and the survival curve was prepared. 48 rats were divided into two groups: CTLL / GG / L / G LCZL / G DXM4 group. They were treated as follows: control group (normal control group), normal feeding group (model group), normal saline 1ml / dL / G LCZ group (prevention group), probiotic powder suspension 2 脳 109 CFU / d and dexamethasone DXM group (dexamethasone group) by intraperitoneal injection of dexamethasone 10 mg 路kg ~ (-1) 路kg ~ (-1), the control group (n = 10) was given intraperitoneal injection of dexamethasone 10 mg / g 路kg ~ (-1). The rats in each group were fed with basic feed for 31 days and fasted for 12 hours. The rats in group B were injected intraperitoneally with LPSN 50 mg / kg and D-GalN 300 mg / kg. The rats were killed 16 hours after treatment. The levels of plasma alanine aminotransferase (alt), aspartate aminotransferase (AST), tumor necrosis factor (TNF- 伪), TLR4, peroxisome proliferator activator receptor-纬 -PPAR 纬 no in liver tissue were measured. The expression of phosphorylated nuclear factor inhibitor protein (p-I 魏 B) PPARYTLR4 in liver tissue was determined. In addition, the pathological changes of liver sections were observed. It was found that the survival rate of group C was lower than that of group A and group D, but significantly higher than that of group B in the survival curve. The results showed that probiotic L.casei Zhang could protect the liver of rats to some extent. The expression of TLR4p-I 魏 B in liver tissue of rats with reduced liver injury was significantly lower than that in L- P / L / G LCZ group and L / G DXM group. The expression of MDA in liver tissue was significantly lower than that in L / G group (P 0.05% L / G LCZ group) compared with that in L / G group (P 0.05% G LCZ group) and LG DXM group (P < 0.05 P / P / G DXM group) compared with that in L / G group (P < 0.05% P < 0.05 P / L / G LCZ group). The expression of MDA in liver tissue was significantly lower than that in L / G group (P < 0.05 L / G LCZ group). The activity of SOD in P0. 05 was significantly higher than that in L- / G group. Probiotics intervention can reduce the acute hepatocyte injury induced by endotoxin. Therefore, the probiotic L.casei Zhang has a good protective effect on endotoxin induced liver injury and can be used in the development of functional food.
