氧浓度的变化对大鼠骨髓间充质干细胞粘附能力的影响

【摘要】:背景间充质干细胞是具有多向分化潜能的非造血干细胞,已广泛应用于干细胞移植、组织工程和器官移植免疫治疗等方面,特别是成为构建组织工程血管中重要的种子细胞[1],但其具体机制不完全清楚。 目的探讨在氧浓度改变对大鼠骨髓间充质干细胞粘附能力的影响。 方法采用SD大鼠20只,年龄6-8周、雌雄不限、体重100g-120g,获取其股骨和胫骨骨髓细胞悬液,贴壁法分离骨髓间充质干细胞。流式细胞鉴定实验细胞CD54、 CD29、CD90、CD45表达,骨髓间充质干细胞分别在分为低氧组(1%O2)和常氧组(20%O2)培养0-7天。根据细胞培养和冲涮时氧浓度,细胞粘附实验分组为20%O2培养20%O2粘附冲涮组(A组)、1%O2培养1%O2粘附冲涮组(B组)、20%O2培养1%O2粘附冲涮组(C组)和1%O2培养20%O2粘附冲涮组(D组),比较不同氧浓度变化条件下细胞残留率。研究不同氧浓度变化对间充质干细胞表达HIF-1a,Integrin beta1,VCAM1和ICAM1的影响,实验分为4组:1组:恒定20%O2培养;2组:恒定1%O2培养;3组:20%O2培养后于1%O2继续培养2h;4组:1%O2培养后20%O2继续培养2h。 结果流式细胞检测99%以上实验细胞表达CD54+、CD29+、CD90+,1%实验细胞表达CD45+,证实细胞为骨髓间充质干细胞。骨髓间充质干细胞在1%O2环境下(A组和B组)粘附能力增强,,在20%O2环境下(C组和B组)粘附能力减弱(P0.05);Integrin beta1,HIF-1a表达水平在1%O2环境下(2组和3组)增强(P0.05)。 结论本研究发现骨髓间充质干细胞随着粘附时间越长,细胞粘附能力也逐渐增强,经过低氧处理细胞的粘附力提高时间快于常氧环境
[Abstract]:Background Mesenchymal stem cells are non-hematopoietic stem cells with multiple differentiation potential. They have been widely used in stem cell transplantation, tissue engineering and organ transplantation immunotherapy. Especially as an important seed cell in the construction of tissue engineering blood vessel, but its mechanism is not completely clear. Objective to investigate the effect of oxygen concentration on the adhesion of rat bone marrow mesenchymal stem cells (BMSCs). Methods Bone marrow mesenchymal stem cells (BMSCs) were obtained from 20 SD rats, aged 6-8 weeks, weighing 100g-120g, and bone marrow mesenchymal stem cells (BMSCs) were isolated by adherent method. CD54, CD29 CD90 and CD45 expression were detected by flow cytometry. Bone marrow mesenchymal stem cells were cultured for 0-7 days in hypoxia group and normoxic group respectively. The cell adhesion test was divided into two groups: 20O 2 culture and 20% O 2 adhesion scouring group A: O 2 group 1 O 2 culture, 2 O 2 adhesion flushing group B group (n = 1) and 1 O 2 culture group 1 O 2 adhesion rinse group C group (C group) and 1 O 2 culture 20 times O 2 adhesion rinse group D group, the cells were compared under different oxygen concentration change conditions. To study the effects of different oxygen concentrations on the expression of HIF-1aHIF-1 integrin beta1 VCAM1 and ICAM1 in mesenchymal stem cells. The experiment was divided into 4 groups: group 1: group 2: group 2: group 2: group 2: group 2: group 2: group 3: group 1: 2: 2 after 2 h culture, group 1: 4, group 1: 1, group 2: 2, group 2: 2, group 2: 1, group 2: 1, group 2: 1, group 1: 1, group 2: 2: 2. Results flow cytometry was used to detect the expression of CD45 in more than 99% experimental cells, which confirmed that the cells were bone marrow mesenchymal stem cells. The adhesiveness of bone marrow mesenchymal stem cells was enhanced in group A and B under 1O 2 environment. The adhesion ability of group C and group B was decreased under the condition of 20O 2. The expression of P0.05 integrin beta1a HIF-1a was enhanced in group 2 and group 3 (P 0.05). Conclusion in this study, the adhesion ability of bone marrow mesenchymal stem cells increased with the increase of adhesion time, and the adhesion of cells treated with hypoxia was faster than that of normoxic environment.
